RNA splicing enables the functional adaptation of cells to changing contexts. Impaired splicing has been associated with diseases, including retinitis pigmentosa, but the underlying molecular mechanisms and cellular responses remain poorly understood. In this work, we report that deficiency of ubiquitin-specific protease 39 (USP39) in human cell lines, zebrafish larvae, and mice led to impaired spliceosome assembly and a cytotoxic splicing profile characterized by the use of cryptic 5' splice sites.

  • Nager syndrome is a rare condition caused by mutations in the SF3B4 gene, leading to facial and limb malformations.
  • Research in fish models lacking SF3B4 showed defects in craniofacial structure and issues with cell signaling due to reduced levels of FGF8.
  • The study found that introducing FGF8 could help fix these defects, suggesting potential new treatments for Nager syndrome by targeting FGF8 pathways.

Pain on the dorsal side of the wrist is a common clinical presentation, comparable to pain experienced on the ulnar and radial aspects of the wrist. The dorsal wrist region has distinct anatomical features and is associated with a wide spectrum of pathologies, including conditions affecting the bones, cartilage, ligaments, and tendons. Accurate diagnosis often depends on imaging techniques such as radiographs and ultrasound, with a growing trend towards the use of magnetic resonance imaging (MRI) for more detailed assessment of complex cases.

This Letter presents results of a search for the mixing of a sub-eV sterile neutrino with three active neutrinos based on the full data sample of the Daya Bay Reactor Neutrino Experiment, collected during 3158 days of detector operation, which contains 5.55×10^{6} reactor ν[over ¯]_{e} candidates identified as inverse beta-decay interactions followed by neutron capture on gadolinium. The analysis benefits from a doubling of the statistics of our previous result and from improvements of several important systematic uncertainties.

  • - Lenacapavir (LEN) is a long-acting injectable drug approved for treating HIV-1, notable for its dual atropisomeric form that maintains a stable ratio in the bloodstream despite rapid interconversion.
  • - The drug shows low systemic clearance in humans and animals, with a primary elimination route through fecal excretion rather than urine, while its concentration remains stable in plasma.
  • - The study reveals that LEN does not enter the brain due to its interaction with P-glycoprotein and emphasizes the significance of intestinal excretion (IE) for drugs that are metabolically stable and eliminated slowly.

The organ-specific toxicity resulting from microplastic (MP) exposure has been extensively explored, particularly concerning the gut, liver, testis, and lung. However, under natural conditions, these effects are not restricted to specific organs or tissues. Investigating whether MP exposure presents a systemic threat to an entire organism, impacting factors such as lifespan, sleep, and fecundity, is essential.

Background: The pathogenesis of congenital diaphragmatic hernia (CDH) depends on multiple factors. Activation of the DNA-sensing cyclic-GMP-AMP-synthase (cGAS) and Stimulator-of-Interferon-Genes (STING) pathway by double-stranded DNA (dsDNA) links environmental stimuli and inflammation. We hypothesized that nitrofen exposure alters cGAS and STING in human bronchial epithelial cells and fetal rat lungs.

There has been increased interest in practical methods for integrative analysis of data from multiple studies or samples, and using factor scores to represent constructs has become a popular and practical alternative to latent variable models with all individual items. Although researchers are aware that scores representing the same construct should be on a similar metric across samples-namely they should be measurement invariant-for integrative data analysis, the methodological literature is unclear whether factor scores would satisfy such a requirement. In this note, we show that even when researchers successfully calibrate the latent factors to the same metric across samples, factor scores-which are estimates of the latent factors but not the factors themselves-may not be measurement invariant.
