The effect of Ca2+ on morphophysiological parameters of calluses of wheat Triticum aestovum L., the level of active oxygen species, and the activity of oxalate oxidase, peroxidase, and catalase is investigated in the case of infestation with the fungus Triticum aestivum causing ball smut. The concentration of O2-, H2O2, and activity of oxidoreductases (oxalate oxidase, peroxidase, and catalase) depends on the content of Ca2+ ions in the culture medium of calluses.

Stability of angiotensin-converting enzyme was studied as a dependence on the zinc-ions concentrations brining in the apo-enzyme. Our data were discussed in the terms of a set of initial permissible conformation conditions of a protein (conformation distribution). Apo-enzyme was shown to be able to the radiation activation that is disappearing in the presence of even 10(-6 )mol/l of the zinc-ions.

The effect of salicylic acid (SA) on oxalate oxidase and peroxidase activities and hydrogen peroxide (H2O2) production in leaf cells has been studied in wheat of the susceptible cultivar Zhnitsa infected by the fungus Septoria nodorum, a pathogen of wheat leaf blotch. The results show that fungal hyphae spread into interstices between mesophyll cells and that infected tissues contain H2O2. Treatment with SA results in enhanced H2O2 production in mesophyll cells, which is due to activation of oxalate oxidase and peroxidase in the cell wall.

A study was made of the influence of bisol 2 and baitan compounds on morphogenesis and defence response of wheat calluse cells infected with bunt agent in associated with oxalate oxidase activation. After introduction of bisol and baitan into cultivation medium, dense area with meristema zones, germs of shoots and rhizoids appeared on non-morphogenic calluses, which correlated with enzyme activation. Parenchyma-like cells, generating hydrogen peroxide, were seen in the site of pathogen penetration under infestation, but were never revealed in control.

We studied the effect of various fungal phytopathogens and exogenous phytohormones on the rate of o-phenylenediamine oxidation mediated by oxalate oxidase, which is one of protective proteins of wheat. The pattern of changes in o-phenylenediamine oxidation rate depended on pathogen type, cultivar resistance, and phytohormone class. This reaction is proposed as a model reaction for studying plant response to infection and effects of various physiologically active compounds.

We have studied the influence of various concentrations of chitooligosaccharides (CO) on hydrogen peroxide (H2O2) generation, with the involvement of oxalate oxidase (OO), in rhizoids and in zones of hard brand pathogen penetration, as well as on oxalate oxidase activity in wheat calluses. In the control group, diaminobenzidin (DAB)-staining was typical of 30% of peripheral rhizoid cells, which provided their resistance during infection. In the zone of fungus penetration, the appearance of DAB-stained parenchyma-like cells was observed.

The dose response of soluble and membrane forms of angiotensin-converting enzyme to gamma-irradiation is investigated at different pH values of the medium and at different concentrations of acetate-phosphate buffer. Membrane form of the enzyme is more stable shows principally other conformational equilibrium than the soluble form. A method for determination of oxidation of phenolic compounds by intact wheat seedlings using o-phenylenediamine (OPD) was developed. The reaction is initiated by the addition of oxalic acid to the incubation medium. It is suggested that an endogenous peroxidase and hydrogen peroxide formed during oxidation of oxalic acid by endogenous oxalate oxidase are involved in OPD oxidation.

Radiation activation of angiotensin-converting enzyme (respect to Cbz-Phe-His-Leu as substrate) was obtained at the gamma (137Cs, t(irr) = 10s-2h, D approximately 3 Gy)- and X (plasma foces source, t(irr) = 10(-9)s, Cu-filter, D approximately 2 x 10(-5) Gy)-irradiation. The inactivation of the horseradish peroxidase at the same X-irradiation dose (2 x 10(-5) Gy) took place. Based on the experimental data and on the mathematical model proposed by us we made a conclusion that the special points exist on the dose response curves.
