Influence of Matrigel-overlay on constitutive and inducible expression of nine genes encoding drug-metabolizing enzymes in primary human hepatocytes.

1. Previous studies reported that rat hepatocytes overlaid with extracellular matrix components (Matrigel) maintain the expression and responsiveness of drug-metabolizing enzymes. However, whether Matrigel provides similar advantages in human hepatocytes remains largely uncertain.

Racial differences in rate of decline in bone mass in older men: the Baltimore men's osteoporosis study.

Lactate and base deficit in trauma: does alcohol or drug use impair their predictive accuracy?

Background: Abnormal blood lactate and base deficit (BD) reflect hypoperfusion and have been documented to predict outcome in trauma. Alcohol and drug use may also induce metabolic acidosis in trauma victims, potentially diminishing the predictive accuracy of lactate and BD. We, therefore, sought to examine the effect of alcohol and drug use on the predictive accuracy of admission blood lactate and BD in trauma.

Eyeblink conditioning deficits indicate timing and cerebellar abnormalities in schizophrenia.

Accumulating evidence indicates that individuals with schizophrenia manifest abnormalities in structures (cerebellum and basal ganglia) and neurotransmitter systems (dopamine) linked to internal-timing processes. A single-cue tone delay eyeblink conditioning paradigm comprised of 100 learning and 50 extinction trials was used to examine cerebellar timing circuits in 13 medicated patients with schizophrenia and 13 age- and sex-matched controls. Patients with schizophrenia showed impaired learning of the conditioned response compared to controls and also greater within-subject variability in the timing of their responses.

Lactams as prostanoid receptor ligands. Part 4: 2-Piperidones as selective EP4 receptor agonists.

2-Piperidones were prepared bearing heptanoic acid or a thioether heptanoic acid at the 1-position as well as appropriately substituted at the 6-position to mimic the structure of prostaglandins. The stereochemical purity at the 6-position was determined to be 95% ee for an advanced synthetic intermediate. The 2-piperidones were identified as potent agonists at the EP4 prostanoid receptor.

Phosphine oxide polymer for water-soluble nanoparticles.

A phosphine oxide polymer was developed using bis(dichlorophosphino)ethane and poly(ethylene glycol). This polymer system was used to transfer various nanoparticles from organic solvents to water, retaining their physical properties and reactivities.

Allogenic blood transfusion in the first 24 hours after trauma is associated with increased systemic inflammatory response syndrome (SIRS) and death.

Background: Previous studies have documented that blood transfusion incites a substantial inflammatory response with the systemic release of cytokines. Furthermore, blood transfusion is a significant independent predictor of multiple organ failure in trauma. The objective of this study was to assess the risk of systemic inflammatory response syndrome (SIRS) and intensive care unit (ICU) admission, length of stay (LOS), and mortality in trauma patients who require blood transfusion.

Phytochemical-induced changes in gene expression of carcinogen-metabolizing enzymes in cultured human primary hepatocytes.

The naturally occurring compounds curcumin (CUR), 3,3'-diindolylmethane (DIM), isoxanthohumol (IXN), 8-prenylnaringenin (8PN), phenethyl isothiocyanate (PEITC) and sulforaphane (SFN) protect animals against chemically induced tumours. Putative chemoprotective mechanisms include modulated expression of hepatic biotransformation enzymes.

Modulation of Alzheimer's pathology by cerebro-ventricular grafting of hybridoma cells expressing antibodies against Abeta in vivo.

Accumulation in brain of the beta-amyloid peptide (Abeta) is considered as crucial pathogenic event causing Alzheimer's disease (AD). Anti-Abeta immune therapy is a powerful means for Abeta clearance from the brain. We recently showed that intravenous injections of anti-Abeta antibodies led to reduction, elevation or no change in brain Abeta42 concentrations of an AD mouse model.

Epidemiology of sepsis in patients with traumatic injury.

Objective: To characterize the epidemiology of sepsis in trauma.

Design: Analysis of a prospectively collected administrative database (Pennsylvania trauma registry).

Setting: All trauma centers in the state of Pennsylvania (n = 28)

Patients: All patients (n = 30,303) with blunt or penetrating injury admitted to Pennsylvania trauma centers over a 2-yr period (January 1996-December 1997).

No association of a non-synonymous PLAU polymorphism with Alzheimer's disease and disease-related traits.

A 30 cM broad genomic region on the long arm of chromosome 10 at 80 cM shows significant and consistent linkage with AD and with plasma concentration of the beta-amyloid peptide 1-42 (Abeta42). The PLAU gene, which is involved in the production and degradation of Abeta42, maps to that region and is therefore a strong positional candidate for association with sporadic AD. We analyzed the non-synonymous single nucleotide polymorphism (SNP) rs2227564 in two independent case-control series from Switzerland and Greece and investigated the influence of this SNP on cognition in elderly individuals.

Moving to an assisted living facility: exploring the transitional experience of elderly individuals.

This qualitative study examined the effects of relocating to an assisted living facility in a select sample of elderly individuals. Small group interviews with open-ended discussion allow participants to express feelings about their transitional experi ence and recall situations in their adjustment process. Theme arising from the data included self-motivating factors, ties to the past, independence, affection, and adjustment, which co related with the principles of assisted living.

Children and adolescents with developmental disabilities. The GP's role.

Background: General practitioners play a central role in the health care for families of children with disabilities, but may feel inadequately trained or resourced for this role.

Objective: This article highlights the important issues arising for children and their families as the child grows--from birth through to adolescence and adulthood. It also provides some key resources for the GP, patient and their family during this transition.

Functional MRI predicts post-surgical memory following temporal lobectomy.

Temporal lobectomy is an effective therapy for medically refractory temporal lobe epilepsy (TLE), but may be complicated by amnestic syndromes. Therefore, pre-surgical evaluation to assess the risk/benefit ratio for surgery is required. Intracarotid amobarbital testing (IAT) is currently the most widely used method for assessing pre-surgical memory lateralization, but is relatively invasive.

A dissociation between implicit and explicit verbal memory in left temporal lobe epilepsy.

Purpose: Temporal lobe epilepsy patients are well known to present deficits on explicit verbal memory procedures (e.g., recall, recognition).

Ultrasound diagnosis of deep venous thrombosis.

The ultrasound diagnosis of deep venous thrombosis by an emergency physician is occurring with increased frequency. The examination is simple to perform and, when combined with a clinical pretest probability or D-dimer, can aid in the rapid disposition ofa patient with lower extremity pain and swelling. The technique and findings of the limited lower extremity ultrasound and the data to support its use in the emergency department are discussed.

Male gender is associated with increased risk for postinjury pneumonia.

Nosocomial pneumonia in trauma patients is a significant source of resource utilization and mortality. We have previously described increased rates of pneumonia in male trauma patients in a single institution study. In that study, female trauma patients had a lower incidence of postinjury pneumonia but a higher relative risk for mortality when they did develop pneumonia.

Two Replicable Suppressor Situations in Personality Research.

Suppressor situations occur when the simultaneous inclusion of two predictors improves one or both validities. A common allegation is that suppressor effects rarely replicate and have little substantive import. We present substantive examples from two established research domains to counter this skepticism.

Anti-amyloid activity of neprilysin in plaque-bearing mouse models of Alzheimer's disease.

Abnormally high concentrations of beta-amyloid peptide (Abeta) and amyloid plaque formation in Alzheimer's disease (AD) may be caused either by increased generation or by decreased degradation of Abeta. Therefore, activation of mechanisms that lower brain Abeta levels is considered valuable for AD therapy. Neuronal upregulation of neprilysin (NEP) in young transgenic mice expressing the AD-causing amyloid precursor protein mutations (SwAPP) led to reduction of brain Abeta levels and delayed Abeta plaque deposition.

Show your pride: evidence for a discrete emotion expression.

Three experiments provide converging evidence that pride has a distinct, recognizable expression. Experiment 1 showed that judges can agree in identifying a posed expression as showing pride and can reliably distinguish pride expressions from expressions of related emotions such as happiness. Experiment 2 showed that judges can identify the pride expression when the task uses an open-ended response format that does not cue them with the label "pride.

The relationship of sociotropy and autonomy to posttraumatic cognitions and PTSD symptomatology in trauma survivors.

The current study examined relationships between sociotropic and autonomous personality styles and posttraumatic stress disorder (PTSD) symptomatology following trauma as well as specific posttraumatic cognitions that have been shown to characterize individuals with PTSD. Self-report data were collected in a sample of 156 college students indicating a history of traumatic experience. Significant relationships were found between symptoms of PTSD and depression and measures of sociotropy, autonomy, and negative posttraumatic beliefs about self and world.
