Article Synopsis
  • The study of metal ions in cell signaling has shifted focus from primarily calcium (Ca) to include other metals like magnesium and d-block metals, suggesting they also play important roles in signal transduction.
  • Fluorescence microscopy is highlighted as a powerful tool for real-time observation of metal transients, helping identify where these ions come from and what molecular targets they affect.
  • The article emphasizes recent advancements in imaging and molecular indicators for studying magnesium, while also addressing ongoing challenges and the broader implications for understanding metal ions in biological processes.

Innovative new crystallographic methods are facilitating structural studies from ever smaller crystals of biological macromolecules. In particular, serial X-ray crystallography and microcrystal electron diffraction (MicroED) have emerged as useful methods for obtaining structural information from crystals on the nanometre to micrometre scale. Despite the utility of these methods, their implementation can often be difficult, as they present many challenges that are not encountered in traditional macromolecular crystallography experiments.
