Article Synopsis
  • The study addresses challenges in interpreting large datasets like TCGA, aiming to identify gene expression profiles linked to patient prognosis.
  • LOCC (Luo's Optimization Categorization Curve) is introduced as a new method for visualizing gene expression against patient outcomes, showing superior predictive capabilities over traditional methods like ROC curves.
  • The analysis revealed specific gene associations with prognosis in hepatocellular carcinoma, positioning LOCC as a valuable tool for cancer research and gene signature evaluation.

Synthetic oleanane triterpenoids (SOTs) are small molecules with broad anticancer properties. A recently developed SOT, 1-[2-cyano-3,12-dioxooleana-1,9(11)-dien-28-oyl]-4(-pyridin-2-yl)-1H-imidazole (CDDO-2P-Im or '2P-Im'), exhibits enhanced activity and improved pharmacokinetics over CDDO-Im, a previous generation SOT. However, the mechanisms leading to these properties are not defined.

A common mechanism in inherited ataxia is a vulnerability of DNA damage. Spinocerebellar ataxia type 7 (SCA7) is a CAG-polyglutamine-repeat disorder characterized by cerebellar and retinal degeneration. Polyglutamine-expanded ataxin-7 protein incorporates into STAGA co-activator complex and interferes with transcription by altering histone acetylation.
