Article Synopsis
  • About 10%-30% of Hodgkin's lymphoma patients experience relapse or treatment resistance after first-line therapy, making subsequent treatment difficult, particularly after failure of Brentuximab vedotin (BV) and immune checkpoint inhibitors (ICIs).
  • A study analyzed 19 patients with relapsed/refractory Hodgkin's lymphoma who underwent radiotherapy after BV or ICI failure, finding a 100% overall response rate and a median duration of response of 17.2 months.
  • The 1-year progression-free survival and overall survival rates were 84.4% and 100%, respectively, with no serious adverse events reported, demonstrating that radiotherapy is both effective and safe for this patient group.

Objective: YAP1 plays a dual role as an oncogene and tumor suppressor gene in several tumors; differentiating between these roles may depend on the YAP1 phosphorylation pattern. The specific function of YAP1 in B cell acute lymphoblastic leukemia (B-ALL), however, is currently unclear. Thus, in the present study, the role of YAP1 in B-ALL was investigated using relevant cell lines and patient datasets.

Article Synopsis
  • Radiotherapy (RT) is a local treatment option for patients with refractory or relapsed aggressive B-cell lymphomas, but the effectiveness of hypofractionated RT had not been previously confirmed.
  • A study analyzed 30 patients who received hypofractionated RT, showing a high objective response rate (90%) and complete response rate (80%), with promising one-year overall survival (81.8%) and progression-free survival (66.3%).
  • The findings indicate that hypofractionated RT is both effective and tolerable, particularly for patients with localized residual disease, as no serious side effects were reported.

Background: Castleman disease (CD) is a group of rare and heterogenous lymphoproliferative disorders including unicentric CD (UCD), human herpesvirus-8(HHV-8)-associated multicentric CD (HHV8-MCD), and HHV-8-negative/idiopathic multicentric CD (iMCD). Knowledge of CD mainly comes from case series or retrospective studies, but the inclusion criteria of these studies vary because the Castleman Disease Collaborative Network (CDCN) diagnostic criteria for iMCD and UCD were not available until 2017 and 2020, respectively. Further, these criteria and guidelines have not been systematically evaluated.

Background: Drug-resistant microbes pose a global health concern, requiring the urgent development of effective antibacterial agents and strategies in clinical practice. Therefore, there is an urgent need to explore novel antibacterial materials to effectively eliminate bacteria. The synthesis of quaternary phosphonium salt in haloargentate systems, wherein the phosphorus atom is represented in a cationic form, is a possible strategy for the development of antibacterial materials.

Article Synopsis
  • The study examined the link between body mass index (BMI) and the prognosis of mantle cell lymphoma (MCL) in 108 patients, revealing that patients with a higher BMI had better progression-free survival (PFS) and overall survival (OS) rates compared to those with a lower BMI.
  • Results indicated that high BMI (≥ 24 kg/m²) was associated with significantly better 5-year PFS (64.6%) and OS (68.7%) compared to low BMI (31.9% PFS and 47.0% OS), also identifying BMI as an independent predictor for both survival rates.
  • The study concluded that BMI plays a crucial role in predicting outcomes

Background: Limited evidence supports the omission of routine bone marrow (BM) examination (biopsy and aspiration) in patients with nasal-type extranodal NK/T-cell lymphoma (ENKTCL). This study was aimed at assessing whether BM examination provides valuable information for positron emission tomography/computed tomography (PET/CT)-based staging in this patient population.

Patients And Methods: Patients newly diagnosed with ENKTCL who underwent initial staging with both PET/CT and BM examination between 2013 and 2020 were retrospectively identified in two Chinese institutions.

Objective: Circulating tumor DNA (ctDNA) is emerging as a versatile biomarker for noninvasive genotyping and response monitoring in specific B-cell lymphomas; however, few studies have been conducted to explore ctDNA-based mutation profiling across non-Hodgkin lymphomas (NHLs) and genomic changes after initiation of chemotherapy.

Methods: A targeted sequencing of 362 genes was performed to detect the mutation profiles in paired blood and tissue samples from 42 NHL patients. Genomic alterations were explored in 11 diffuse large B-cell lymphoma (DLBCL) patients using paired blood samples collected pre- and post-R-CHOP chemotherapy.

Background: Mounting studies have sought to identify novel mutation biomarkers having diagnostic and prognostic potentials. Nevertheless, the understanding of the mutated pathways related to development and prognosis of B-cell lymphoma is still lacking. We aimed to comprehensively analyze the mutation alterations in genes of canonical signaling pathways and their impacts on the clinic outcomes of patients with B-cell lymphoma.

Myosin light chain 9 (MYL9) is a regulatory light chain of myosin, which plays an important role in various biological processes including cell contraction, proliferation and invasion. MYL9 expresses abnormally in several malignancies including lung cancer, breast cancer, prostate cancer, malignant melanoma and others, which is closely related to the poor prognosis, but the clinical significance for its expression varies with different types of cancer tissues. Further elucidating the molecular mechanism of MYL9 in various types of malignant tumor metastasis is of great significance for cancer prevention and treatment.

Diffuse large B-cell lymphoma (DLBCL) is the most common adult non-Hodgkin lymphoma (NHL) and is highly invasive, with a poor prognosis. The main clinical treatment for DLBCL involves chemotherapy or a combination of chemotherapy and targeted drugs. CD56 expression is considered as an indicator of poor prognosis in patients with acute myeloid leukemia and anaplastic large cell lymphoma; however, its role in DLBCL remains unclear.

Background: The International Prognostic Index (IPI) scoring system is the most widely used prognostic tool for diffuse large B-cell lymphoma (DLBCL); however, it fails to consistently identify patients with poor outcomes. This retrospective study was undertaken to confirm the clinical value of a new prognostic score and compare it with the IPI.

Methods: The aim of this single-center study was to confirm the clinical value of a new prognostic score and its association with various clinical features, disease progression, and death in 70 patients with DLBCL who had undergone at least 6 cycles of chemotherapy.
