Introduction: Alzheimer's disease (AD) is the most common form of dementia in the elderly. Given that AD neuropathology begins decades before symptoms, there is a dire need for effective screening tools for early detection of AD to facilitate early intervention.

Methods: Here, we used tree-based and deep learning methods to train polyomic prediction models for AD affection status and age at onset, employing genomic, proteomic, metabolomic, and drug use data from UK Biobank.

Article Synopsis
  • Alzheimer's disease (AD) is the most common form of dementia, and while many genetic factors have been found, research has mostly focused on people of European descent.
  • This study analyzed whole genome sequencing data from a diverse group of participants (about 49,000 AD cases and 383,000 controls) to identify new genetic loci associated with AD.
  • The researchers discovered 16 novel loci linked to clinically-diagnosed AD and AD-by-proxy, emphasizing the need for more diverse studies in genetic research.

Patients with coccidioidal meningitis require lifelong antifungal therapy. Cumulative toxicity and lack of antifungal efficacy require salvage therapy in the treatment of some patients. In a retrospective review of nine patients with coccidioidal meningitis treated with isavuconazole, successful therapy was seen in three patients and stable disease was confirmed in six patients.
