Hydrolysis-resistant RNA-peptide conjugates that mimic peptidyl-tRNAs are often required for structural and functional studies of protein synthesis at the ribosome. These conjugates can be synthesized by solid-phase chemical synthesis, which allows maximum flexibility in both the peptide and RNA sequence. The commonly used strategy is based on (3'-N-aminoacyl)-3'-amino-3'-deoxyadenosine solid supports, which already contain the first C-terminal amino acid of the target peptidyl chain.

Current blood coagulation models consider the interactions between blood, the vessel wall, and other tissues that expose tissue factor (TF), the main initiator of coagulation. A potential role of body fluids other than blood is generally not considered. In this review, we summarize the evidence that body fluids such as mother's milk saliva, urine, semen, and amniotic fluid trigger coagulation.

Article Synopsis
  • Human saliva contains extracellular vesicles (EVs) that can activate blood coagulation through the presence of extrinsic tenase complexes, which include tissue factor and activated factor VII.
  • Individuals with severe hemophilia A, who lack FVIII, have salivary EVs that can compensate for their condition by generating FXa, leading to infrequent oropharyngeal bleeding.
  • In contrast, people with severe FVII deficiency lack these functional extrinsic tenase complexes in their saliva, resulting in more frequent oropharyngeal bleedings; however, adding recombinant FVIIa can restore coagulation potential in their saliva.

Article Synopsis
  • The study investigates the use of methyl jasmonate, a plant hormone, to enhance resistance in corn seeds against seed corn maggot pests, focusing on how different application methods affect pest behavior and plant growth.
  • Results showed that soaking seeds in a 0.2 mM solution of methyl jasmonate significantly reduced the preference of adult flies to lay eggs on treated seeds, while having minimal impact on seedling growth.
  • Overall, treating corn seeds with methyl jasmonate could effectively deter pest infestation during critical growth periods, suggesting a viable pest management strategy without severely hindering plant development.

Background: Scientific and clinical interest in extracellular vesicles (EVs) is growing. EVs that expose tissue factor (TF) bind factor VII/VIIa and can trigger coagulation. Highly procoagulant TF-exposing EVs are detectable in the circulation in various diseases, such as sepsis, COVID-19, or cancer.

Immune thrombotic thrombocytopenic purpura (iTTP) is a rare, life-threatening autoimmune disorder caused by a disintegrin and metalloproteinase with thrombospondin type 1 motif, member 13 (ADAMTS13) deficiency. Caplacizumab, an anti-von Willebrand factor nanobody, is approved for iTTP treatment, reducing the need for therapeutic plasma exchange (TPE) and improving platelet count recovery and survival. We conducted a retrospective study on 42 acute iTTP cases in Austria and Germany, treated with a modified regimen aimed at avoiding TPE if platelet count increased after the first caplacizumab dose.

Article Synopsis
  • Acute myeloid leukemia (AML) and acute lymphoblastic leukemia (ALL) are serious blood cancers that involve the rapid increase of abnormal white blood cells, and recent research has highlighted BRD4 and MYC as potential targets for new drug therapies.
  • In experiments comparing various BRD4-targeting drugs, including the BET inhibitor JQ1 and BRD4 degraders dBET1 and dBET6, all three were effective at reducing the growth of AML and ALL cells, including resistant leukemic stem cells.
  • Notably, dBET6 outperformed the others by overcoming drug resistance, showing especially promising results when paired with other medications, and it also inhibited the expression of the resistance-related PD-L1 antigen in

Objectives: Clinical examination alone cannot reliably rule out significant traumatic abdominal injury. Computed tomography (CT) has become the primary method for evaluating blunt abdominal trauma and clinicians rely heavily on it to rule out abdominal injury. Ultrasound examination may miss significant abdominal injury particularly in stable patients.

Introduction: Controlled randomized trials, molecular analytics, and guideline recommendations have so far been irreplaceable tools to ensure appropriate treatment and decision-making for physicians and patients. Individual patient models are increasingly complementing these methods, particularly in the case of advanced cancers, rare cancers, and cancers of unknown primary (CUP), as in these cases comprehensive clinical evidence is unavailable, often resulting in poor treatment success, even after stratification.

Case Presentation: Here we report a 53-year-old patient with CUP with axillary lymph node metastases for whom patient-derived 3D (PD3D) tumor organoids successfully guided personalized treatment.

In 2007, the ASSO-LM1 trial, a multicenter prospective study, was initiated to investigate the resectability (R0) rate following preoperative combination therapy with XELOX and bevacizumab in patients with potentially resectable colorectal liver metastases. Six cycles of systemic therapy were administered preoperatively, although the sixth cycle did not include bevacizumab, resulting in 5 weeks between the last bevacizumab dose and surgery. Treatment with bevacizumab plus XELOX was restarted for another six cycles postoperatively.

Soil legacy influences plant interactions with antagonists and below-ground mutualists. Plant-antagonist interactions can jeopardize plant-pollinator interactions, while soil mutualists can enhance plant-pollinator interactions. This suggests that soil legacy, either directly or mediated through plant symbionts, affects pollinators.

Venous thromboembolism (VTE) is a common complication in patients with cancer. Data on the role of natural inhibitors of coagulation for occurrence of cancer-associated VTE are limited, thus, we investigated the association of tissue factor pathway inhibitor (TFPI) with risk of VTE and all-cause mortality in patients with cancer. Total TFPI antigen levels were measured with a commercially available enzyme-linked immunosorbant assay in patients included in the Vienna Cancer and Thrombosis Study, a prospective observational cohort study with the primary outcome VTE.

Biliary tract cancers are rare cancers with poor prognosis due to a lack of therapeutic options, especially after the failure of first-line systemic treatment. Targeted treatments for this clinical situation are promising and have entered clinical practice. We aimed to describe the overall survival of matched targeted treatment after first-line treatment in patients with biliary tract cancers in an Austrian real-world multicenter cohort.

Pancreatic cancer is one of the most prothrombotic malignancies. Plasma clot properties may be altered in patients with pancreatic cancer, and circulating tissue factor (TF) may play an important role. We applied a modified plasma clot formation assay (only CaCl and phospholipids were added to initiate clotting) and a standard clotting assay (lipidated TF was also added) to investigate whether plasma clot properties are altered in pancreatic cancer patients (n = 40, 23 female) compared to sex-matched healthy controls.

Hypothalamic gliosis associated with high-fat diet (HFD) feeding increases susceptibility to hyperphagia and weight gain. However, the body-weight-independent contribution of microglia to glucose regulation has not been determined. Here, we show that reducing microglial nuclear factor κB (NF-κB) signaling via cell-specific IKKβ deletion exacerbates HFD-induced glucose intolerance despite reducing body weight and adiposity.
