Suspended sediment concentration (SSC) plays a pivotal role in shaping coastal dynamics, impacting terrestrial and marine ecosystems. This study employed the hydrodynamic model MIKE21 to simulate hydrological runoff and sediment transport within the Mekong River's fluvial-marine continuum, the longest river in Southeast Asia currently challenged with escalating anthropogenic pressures and sea-level rise. By strategically selecting hourly observed data from various locations (river channel, coastal estuary) and periods (dry and rainy seasons) for model calibration and validation, we demonstrated the robust performance of the model simulation of both water levels (RMSE: 0.

Lung fibrosis, characterized by chronic and progressive scarring, has no cure. Hallmarks are the accumulation of myofibroblasts and extracellular matrix, as well as vascular remodeling. The crosstalk between myofibroblasts and vasculature is poorly understood, with conflicting reports on whether angiogenesis and vessel density are increased or decreased in lung fibrosis.

Epidermal growth factor receptor (EGFR) tyrosine kinase inhibitors are commonly used to treat non-small cell lung cancers with EGFR mutations, but drug resistance often emerges. Intratumor heterogeneity is a known cause of targeted therapy resistance and is considered a major factor in treatment failure. This study identifies clones of EGFR-mutant non-small cell lung tumors expressing low levels of both wild-type and mutant EGFR protein.

The generation of lung epithelial cells through the directed differentiation of human pluripotent stem cells (hPSCs) in vitro provides a platform to model both embryonic lung development and adult airway disease. Here, we describe a robust differentiation protocol that closely recapitulates human embryonic lung development. Differentiating cells progress through obligate intermediate stages, beginning with definitive endoderm formation and then patterning into anterior foregut endoderm that yields lung progenitors (LPs) with extended culture.

Over the last decade, our knowledge of colorectal serrated polyps and lesions has significantly improved due to numerous studies on this group of precursor lesions. Serrated lesions were misleading as benign before 2010, but they are currently reclassified as precancerous lesions that contribute to 30% of colorectal cancer through the serrated neoplasia pathway. The World Health Organization updated the classification for serrated lesions and polyps of the colon and rectum in 2019, which is more concise and applicable in daily practice.

Hydrogels have emerged as potential materials for bone grafting, thanks to their biocompatibility, biodegradation, and flexibility in filling irregular bone defects. In this study, we fabricated a novel NAH hydrogel system, composed of N,O-carboxymethyl chitosan (NOCC), aldehyde hyaluronic acid (AHA), and hydroxyapatite (HAp). To improve the mechanical strength of the fabricated hydrogel, a porous polycaprolactone (PCL) matrix was synthesized and used as a three-dimensional (3D) support template for NAH hydrogel loading, forming a novel PCL/NAH hybrid scaffold.

Article Synopsis
  • A 2.5-month-old girl was diagnosed with methylmalonic acidemia (MMA) due to a deficiency in transcobalamin II, affecting vitamin B12 metabolism, which resulted in failure to thrive and severe low blood cell counts (pancytopenia).
  • She developed opportunistic Cytomegalovirus infections and pneumonia, leading to severe respiratory distress syndrome (ARDS) and immune reconstitution inflammatory syndrome (IRIS) after starting vitamin B12 treatment.
  • In treating children with lung-related ARDS, doctors should not wait for normal lymphocyte counts to perform necessary invasive procedures for identifying infections, and quick high-dose corticosteroid treatment is crucial for managing inflammation linked to IRIS.

  • - Small-cell lung cancer (SCLC) is a highly lethal form of lung cancer that initially responds well to chemotherapy but becomes resistant at relapse.
  • - The study found that the expression of the TREX1 gene is significantly increased in chemoresistant SCLC, indicating its role in the tumor's survival.
  • - Inhibiting TREX1 may activate the immune response and make resistant SCLC cells more sensitive to chemotherapy, suggesting it could be a novel therapeutic target for improving treatment outcomes.

Article Synopsis
  • - The study aimed to evaluate the effectiveness of combining comparative genomic hybridization and single-nucleotide polymorphism (CGH/SNP) analyses for risk stratification in pediatric acute lymphoblastic leukemia (ALL) compared to traditional cytogenetic methods.
  • - Researchers analyzed data from 135 patients aged 1-18 diagnosed with ALL, finding that CGH/SNP had a significantly lower failure rate and faster result turnaround (5.8 days) than conventional karyotyping (10.7 days).
  • - CGH/SNP detected crucial gene deletions, particularly ETV6, which was associated with better event-free survival, indicating that CGH/SNP could enhance diagnostic accuracy and prognostic evaluation in pediatric ALL.

Philadelphia chromosome-like acute lymphoblastic leukemia (Ph-like ALL) represents a high-risk B-lineage ALL subtype characterized by adverse clinical features and poor relapse-free survival despite risk-adapted multiagent chemotherapy regimens. The advent of next-generation sequencing has unraveled the diversity of kinase-activating genetic drivers in Ph-like ALL that are potentially amenable to personalized molecularly-targeted therapies. Based upon robust preclinical data and promising case series of clinical activity of tyrosine kinase inhibitor (TKI)-based treatment in adults and children with relevant genetic Ph-like ALL subtypes, several clinical trials have investigated the efficacy of JAK- or ABL-directed TKIs in cytokine receptor-like factor 2 (CRLF2)/JAK pathway-mutant or ABL-class Ph-like ALL, respectively.

Neoantigen delivery using extracellular vesicles (EVs) has gained extensive interest in recent years. EVs derived from tumour cells or immune cells have been used to deliver tumour antigens or antitumor stimulation signals. However, potential DNA contamination from the host cell and the cost of large-scale EV production hinder their therapeutic applications in clinical settings.

One of the major hurdles that has hindered the success of chimeric antigen receptor (CAR) T cell therapies against solid tumors is on-target off-tumor (OTOT) toxicity due to sharing of the same epitopes on normal tissues. To elevate the safety profile of CAR-T cells, an affinity/avidity fine-tuned CAR was designed enabling CAR-T cell activation only in the presence of a highly expressed tumor associated antigen (TAA) but not when recognizing the same antigen at a physiological level on healthy cells. Using direct stochastic optical reconstruction microscopy (dSTORM) which provides single-molecule resolution, and flow cytometry, we identified high carbonic anhydrase IX (CAIX) density on clear cell renal cell carcinoma (ccRCC) patient samples and low-density expression on healthy bile duct tissues.
