Fluorescence microscopy has revolutionized biological research by enabling the visualization of subcellular structures at high resolution. With the increasing complexity and volume of microscopy data, there is a growing need for automated image analysis to ensure efficient and consistent interpretation. In this study, we introduce PunctaFinder, a novel Python-based algorithm designed to detect puncta, small bright spots, in raw fluorescence microscopy images without image denoising or signal enhancement steps.

In the unicellular eukaryote Saccharomyces cerevisiae, Cln3-cyclin-dependent kinase activity enables Start, the irreversible commitment to the cell division cycle. However, the concentration of Cln3 has been paradoxically considered to remain constant during G1, due to the presumed scaling of its production rate with cell size dynamics. Measuring metabolic and biosynthetic activity during cell cycle progression in single cells, we found that cells exhibit pulses in their protein production rate.

To acquire more insight into the results of treatment versus the "natural" course of glomus tumors, we studied the clinical data of 108 patients, in 58 of whom the disease was hereditary. During a period of 32 years (1956 to 1988), 175 tumors were diagnosed: 52 glomus jugulotympanic tumors, 32 vagal body tumors, and 91 carotid body tumors. The results of radical surgical treatment were disappointing for tumors located at the skull base, ie, nonradical in 59% (n = 23) of the cases, but very good for the carotid body tumors, for which 96% (n = 68) radical excision was achieved.

Pemphigus vulgaris developed in a 23-year-old woman during the first trimester of pregnancy. In the 33d week she had a stillbirth of a female child who showed extensive skin lesions clinically suggestive of pemphigus. The application of direct immunofluorescence staining to skin tissue sections showed deposition of IgG in the intercellular spaces of the epidermis of both mother and child.

Three cases of contact dermatitis caused by Maneb, manganous-ethylene-bis (dithiocarbamate), are described. Maneb is used as an agricultural fungicide, and is also used in spraying solutions in horticulture and in flower shops. Patch testing with Maneb-related compounds revealed no regular pattern of cross sensitization.
