Rhabdomyosarcoma is the most common soft tissue sarcoma in children and adolescents. Embryonal rhabdomyosarcoma (ERMS), its most common subtype, is a malignant soft tissue tumor with morphologic and immunophenotypic features of embryonic skeletal muscle. The histologic findings in ERMS typically include a range of differentiation in rhabdomyoblasts from primitive to terminally differentiated forms, and the latter become more prominent after chemotherapy-induced cytodifferentiation.

Article Synopsis
  • A CCRF-CEM mutant, referred to as CEM-p, shows resistance to methotrexate due to lower accumulation of its polyglutamate forms.
  • Researchers investigated the underlying mechanisms by analyzing the expression of FPGS and SLC19A1 mRNA, assessing FPGS promoter activity, and sequencing the FPGS promoter and cDNA from both CCRF-CEM and CEM-p cells.
  • The study identified three amino acid changes in FPGS from CEM-p cells that impact ATP binding, glutamate/folate binding, and a critical conserved domain, highlighting the essential role of this domain for FPGS functionality.
