There is a growing number of reports on the occurrence of benzimidazole resistance-associated single nucleotide polymorphisms (SNPs) in the β-tubulin isotype 1 gene of various helminths of veterinary, and public health concerns. However, a comprehensive analysis of their occurrence, and their contributions to conferring benzimidazole resistance among hookworms has yet to be done. The objectives of this systematic review are to summarize and synthesize peer-reviewed evidence on the occurrence of these resistance-associated mutations in hookworms, document their geographical distribution, and assess their contributions to conferring phenotypic resistance.

Article Synopsis
  • This systematic review focused on the characteristics and treatment of ligneous gingivitis (LG) and periodontitis (LP) in patients with plasminogen deficiency (PD), analyzing 17 case reports related to these oral conditions.
  • The findings revealed that 56% of patients had type I PD, with common symptoms including ulceration and bone loss, and the most frequent treatment was conventional periodontal scaling.
  • Limitations in current research create challenges in diagnosing and treating LG/LP, highlighting the need for more comprehensive future studies with detailed treatment descriptions and longer follow-up periods.

Tuft cells have gained substantial attention over the past 10 years due to numerous reports linking them with type 2 immunity and microorganism-sensing capacity in many mucosal tissues. This heightened interest is fuelled by their unique ability to produce an array of biological effector molecules, including IL-25, allergy-related eicosanoids, and the neurotransmitter acetylcholine, enabling downstream responses in diverse cell types. Operating through G protein-coupled receptor-mediated signalling pathways reminiscent of type II taste cells in oral taste buds, tuft cells emerge as chemosensory sentinels that integrate luminal conditions, eliciting appropriate responses in immune, epithelial and neuronal populations.

Background: Epigenetic modifications, such as DNA methylation and histone modifications, are pivotal in regulating gene expression pathways related to inflammation and cancer. While there is substantial research on epigenetic markers in cholangiocarcinoma (CCA), Opisthorchis viverrini-induced cholangiocarcinoma (Ov-CCA) is overlooked as a neglected tropical disease (NTD) with limited representation in the literature. Considering the distinct etiological agent, pathogenic mechanisms, and pathological manifestations, epigenetic research plays a pivotal role in uncovering markers and potential targets related to the cancer-promoting and morbidity-inducing liver fluke parasite prevalent in the Great Mekong Subregion (GMS).

Article Synopsis
  • Hemophilia A is an X-linked disorder caused by a deficiency of coagulation factor VIII, mainly due to mutations in the factor 8 gene, and this study focused on identifying genetic variants in Colombian males with severe hemophilia A.
  • The study analyzed 50 participants and discovered 17 different pathogenic variants, with 70% being truncation variants and 35% being novel mutations not previously reported.
  • A significant finding was that 75% of participants with a history of positive inhibitors had light chain variants, indicating a possible relationship between variant location and inhibitor risk, paving the way for personalized treatment strategies.

Background: A single-dose dengue vaccine that protects individuals across a wide age range and regardless of dengue serostatus is an unmet need. We assessed the safety and efficacy of the live, attenuated, tetravalent Butantan-dengue vaccine (Butantan-DV) in adults, adolescents, and children. We previously reported the primary and secondary efficacy and safety endpoints in the initial 2 years of follow-up.

Objective: This systematic review and meta-analysis sought to assess the published research on mandibular cortical and trabecular bone patterns (TBPs) in individuals with chronic kidney disease (CKD) assessed by radiomorphometric index (RMI) analysis on dental radiographs.

Study Design: The review was conducted in accordance with the Preferred Reporting Items for Systematic Reviews and Meta-Analyses statement. Observational studies that radiographically evaluated cortical and TBPs were analyzed.

Article Synopsis
  • A study in Brazil examined the occurrence and characteristics of oral lesions linked to immune-mediated and autoimmune diseases in patients over a twelve-year period.
  • Out of 3,790 lesions diagnosed, 160 (4.2%) were related to such diseases, mainly affecting middle-aged women, with oral lichen planus, mucous membrane pemphigoid, and pemphigus vulgaris being the most common types.
  • The research highlighted that these lesions often cause pain and require a comprehensive, multidisciplinary approach for management and treatment, with varying times for disease stabilization depending on the condition.

  • The review aimed to analyze the current understanding of medication-related osteonecrosis of the jaw (MRONJ) linked to tooth extractions in patients undergoing bisphosphonate therapy, addressing knowledge gaps for future research.
  • It evaluated a total of 176 studies, with 41.4% focusing on animal models and 58.5% on human subjects, revealing that zoledronic acid was the most studied bisphosphonate, especially with intravenous use.
  • Key findings indicated that the mandible was the primary site of complications, with bone necrosis identified as the most common consequence in both animal (53.4%) and human (22.1%) studies, emphasizing the need for vigilance in patients with a history of

  • - The study examines oral lesions in systemic lupus erythematosus (SLE) patients from Latin America, highlighting their varied characteristics and challenges in diagnosis, especially when lesions appear alone.
  • - In a sample of 70 SLE patients, the majority were white females with an average age of 38.4 years, and the most common oral lesions were ulcers, erosions, and white lesions, often found in the hard/soft palate.
  • - Findings suggest that unusual oral changes may signal underlying systemic issues, emphasizing the need for a multidisciplinary approach and regular oral exams to identify and treat these lesions effectively.

Article Synopsis
  • The review examines the oral health issues faced by heart transplant patients who are on immunosuppressive therapy, focusing on their oral manifestations.
  • A comprehensive literature search identified 402 studies, of which 13 were relevant and included; these studies mainly highlighted periodontal diseases and drug-induced gingival enlargement as common issues.
  • The findings suggest that while drug-induced gingival enlargement is the most frequently reported oral manifestation, other issues like oral cancer and infections were noted less often, indicating variability in research quality and outcomes.

The tuft cell-ILC2 circuit orchestrates rapid type 2 responses upon detecting microbe-derived succinate and luminal helminths. Our findings delineate key mechanistic steps, involving IP3R2 engagement and Ca flux, governing IL-25 production by tuft cells triggered by succinate detection. While IL-17RB plays a pivotal intrinsic role in ILC2 activation, it exerts a regulatory function in tuft cells.
