Esophageal cancer is a highly aggressive disease, and acquired resistance to chemotherapy remains a significant hurdle in its treatment. mtDNA, crucial for cellular energy production, is prone to mutations at a higher rate than nuclear DNA. These mutations can accumulate and disrupt cellular function; however, mtDNA mutations induced by chemotherapy in esophageal cancer remain unexplored.

Background: The association between tumor-infiltrating lymphocytes and tumor immunity has long been recognized. Among T-cell types, CD45RO-positive memory T cells (CD45RO) are reported to correlate with survival in several cancer types, but clinical evidence is lacking in esophageal squamous cell carcinoma (ESCC).

Methods: In surgical specimens from 162 preoperatively untreated patients, immunohistochemistry for CD45RO was performed to evaluate the density of CD45RO in the tumor core (CT) and invasive margin (IM) using an auto-count method.

Article Synopsis
  • Hematoma expansion (HE) occurs in a significant portion of patients with acute intracerebral hemorrhage (ICH), impacting their outcomes; the study focuses on the predictive accuracy of the Black-&-White (B&W) sign in identifying HE.
  • In a multicenter cohort from the PREDICT study, the association between the B&W sign and HE was analyzed, revealing that patients with the B&W sign had a higher frequency of HE and more substantial growth of hematomas compared to those without it.
  • The B&W sign strongly predicts HE, with an adjusted odds ratio of 7.83 for HE and 5.67 for severe HE, indicating that its presence significantly increases the risk of hematoma expansion.

  • Postoperative delirium is a common issue in elderly patients after gastric cancer surgery, affecting about 12.2% of those studied.
  • The study analyzed 255 patients aged 75 and older, linking factors like poor Eastern Cooperative Oncology Group performance status (ECOG-PS), diabetes, and mental decline to higher delirium rates.
  • The researchers found that combining scores from the Mini-Mental State Examination (MMSE) and ECOG-PS can predict delirium effectively, with 80.7% sensitivity and 74.1% specificity.

Article Synopsis
  • Conversion therapy, involving surgery or chemoradiotherapy, is being explored for treating esophageal squamous cell carcinoma (ESCC) patients with distant metastases, which were previously considered incurable.
  • A study reviewed 147 patients who underwent conversion therapy, revealing that most had major complications post-surgery, but overall survival rates were promising, with a 5-year survival rate of 31.7% for patients who responded to treatment.
  • The research concluded that conversion therapy for ESCC with distant metastasis is not only safe but also may lead to better patient outcomes, particularly for those who show a positive response to treatment.

Background/aim: Most clear cell renal cell carcinomas (ccRCCs) have a dysfunctional von Hippel-Lindau tumor suppressor protein (VHL). Hypoxia-inducible factors 1 and 2 alpha (HIF1α and HIF2α) accumulate in ccRCC with dysfunctional VHL and up-regulate the vascular endothelial growth factor (VEGF) pathway and tumor angiogenesis. Recently, pimitespib (PIM), a potent ATP-competitive inhibitor of heat shock protein 90 (HSP90), was developed.
