To know the biological function of the adherens junction associated with histological dedifferentiation, intramuscular invasion, and clinical staging, we studied the expressions of E-cadherin and alpha- and beta-catenin mRNAs in endometrial cancers in comparison with those in normal uterine counterparts. As dedifferentiation, myometrial invasion and staging advanced, the expressions of E-cadherin and beta-catenin mRNAs decreased. This suggests that the function of adhesive molecules in the adherens junction might be disturbed, leading to invasiveness and metastatic potential.

These results suggest that metastatic cancer cells by themselves increase the potential of blood vessel invasion, which can be inhibited by estrogen and progestin administration.

We have explored the mechanism of estrogen-induced growth in human uterine leiomyomas from the aspect of sex hormone-binding globulin (SHBG) exon VII splicing variant mRNA expression using the reverse transcription-polymerase chain reaction-Southern blot and DNA sequencing analyses. The results were obtained by analysis of the missing base pairs corresponding to the entire exon VII, which are considered to encode a portion of the steroid-binding site. This absence replaces 118 amino acids from the carboxy-terminus of SHBG with nine different amino acid residues due to the formation of a new stop codon at residue 334.

We investigated the effects of clarithromycin (CAM) on clinical and endometrial immune responses in chronic intrauterine infection (pyometra). CAM was orally administered in a dose of 200 mg once a day for 4 months (long-term/low-dose treatment) in patients with pyometra. To compare the operative and sexual efficiencies of transabdominal-vulvar and transvulvar techniques for peritoneal vaginoplasty in patients with congenital absence of the vagina, we treated 36 patients by peritoneal vaginoplasty, originally described by Rothman, 31 with the transabdominal-vulvar technique and five with a new transvulvar technique. Both techniques equally gave the patients excellent sexual conditions. The mean blood loss was the mean duration of operation were 278 +/- 268 g and 1 degree 54' +/- 48' in the transabdominal-vulvar technique and 50 +/- 52 g and 58' +/- 11' in the transvulvar technique.
View Article and Find Full Text PDFTo know the potential of growth, invasion and metastasis of endometrial cancer associated with neovascularization, the effects of sex steroids and O-(chloroacetyl-carbamoyl) fumagillol (TNP-470; AGM-1470) on basic fibroblast growth factor (FGF) expression and secretion and its mRNA expression were investigated in well-differentiated endometrial cancer cell line Ishikawa and in undifferentiated endometrial cancer cell line AN3 CA. Basic FGF expression and secretion and its mRNA expression in Ishikawa cells, but not in AN3 CA cells, were increased by estrogen, while progesterone diminished the estrogen-induced increases. TNP-470 reduced the levels regardless of estrogen treatment in AN3 CA cells.
