Unlabelled: Prolactin (PRL) has been detected in the cerebrospinal fluid (CSF) in humans and the absolute level appears to reflect the serum PRL concentration. Because PRL is thought to be involved in the regulation of brain water and electrolyte content attempt has been made to determine CSF and plasma PRL and dopamine (DA) concentrations, osmolality, and sodium level in 21 newborn infants undergoing lumbar punction because of apneic spells, fever, or perinatal asphyxia. Unlabelled: The breast milk prolactin (PRL) has been claimed to play a role in the control of electrolyte composition of the milk. Since metoclopramide has been shown to increase milk production in humans, we have made an attempt to investigate the production, the PRL and sodium concentrations in milk with (group I) and without (group II) maternal metoclopramide treatment (5 days, 30 mg/day). A new cell line (B-25F) obtained from a benign polypoid fibrous lesion of the mucosa of oral cavity is described. The cells at first grew in suspension but after a month of cultivation they began to adhere and subsequently formed a monolayer typical for fibroblastoid cells. Population doubling time both in lower and higher passages was 60 h.

Plasma renin activity and plasma aldosterone concentration were measured by radioimmunoassay in eight newborn infants born vaginally to mothers on indomethacin treatment. Measurements were made in venous cord blood at birth and on the sixth day of life. The results were compared to those obtained in eight healthy control newborn infants of similar gestational age and birth weight.

Simultaneous measurement of plasma renin activity (PRA), plasma aldosterone concentration (PA) and urinary aldosterone excretion (UAE) was made using the RIA method along with determination of Na and K balance in 1-week-old neonates with gestational age of 30-41 weeks (mean 35.9 weeks) and birth weight of 1,160-4,670 g (mean 2,680 g). Full MMTV-Y gene expression can occur in dexamethasone-insulin-prolactin-stimulated cell cultured derived from C57Bl/10 mammary adenocarcinomas induced by syncarcinogenic action of chemical carcinogens (dimethylbenzanthracene), and mammotropic hormones (estrogen and prolactin). The rate of the hormone-stimulated virus production, as determined by biochemical, immunological and electron microscopical methods, was comparable to the level of spontaneous MMTV production by cells of established mouse mammary cancer cultures (CCL-51 and Mm5mt/cl). On the contrary, no virus production has been detected in hormone-stimulated cultures derived from C57Bl/10 mammary tumors induced by chemical carcinogen alone (urethan).

By optimal hormonal treatment the production of exogenously transmitted MMTV can be stimulated in vitro to different degrees, depending on cultivation conditions and origin of tumor cells. Moreover, after appropriate hormonal treatment, endogenous MMTV-Y can be rescued from primary cell cultures derived from dimethyl benzanthracene- and hormone-induced C57BL/10 mouse mammary adenocarcinoma, as determined by reverse transcriptase assay, distribution of 3H-uridine-labelled viral particles, immunofluorescence, and electron microscopy. On the contrary, all attempts to rescue MMTV-Y from cultures derived from urethane-induced C57BL/10 tumors failed.

Murine xenotropic virus, designated 698/X, was recovered by implantation of human lymphosarcoma-derived cells U-698M into nude mice of Giovanella's colony. Treatment of intact avian myeloblastosis virus (AMV) with dimethyl suberimidate dihydrochloride (DMS), a cross-linking agent specific for amino groups, was found to result in progressive cross-linking among viral proteins, as revealed by polyacrylamide gel electrophoresis (PAGE) in the presence of sodium dodecyl sulphate (SDS). Free viral proteins were not cross-linked. The cross-linked protein complex with an apparent molecular weight of 50,000 daltons was studied in detail.

The establishment and cultivation of two human neoplastic cell lines is described. The cell line B-5GT was derived from bone giant cell tumor and B-6FS from poorly differentiated fibrosarcoma. In comparison to the normal skin fibroblasts both cell lines have a potential for "indefinite" multiplication in vitro and they exhibit growth properties which are associated with malignant transformation.

A continuous cell line, RBA-BC, has been derived from the buffy coat of heparinized blood of a Sprague--Dawley rat with developed RBA myeloblastosis. Separations of myeloblasts from the RBA-BC cell line was accomplished by velocity sedimentation in Verografin density gradients. In contrast to the B77 virus, its B77(RBI) rat variant continuously produced by the rat RBI sarcoma cells in vivo and in vitro, efficiently induced progressively growing tumors in hamsters and transformed hamster embryo cells in vitro. Both, induced tumors and transformed cell cultures showed production of infectious virus and presence of the avian gs-antigen. Clonal analysis of the established hamster sarcoma cell line RBHtc brought evidence that despite the quantitative differences in the virus production and gs-antigen content among various clonal populations, full expression of the B77(RBI) virus genome does occur in each cell of the virus-induced hamster sarcoma.
