Background: Direct skeletal fixation, a surgical technique enabling the attachment of an external prosthesis directly to the bone through a percutaneous implant, offers an enticing solution for patients with lower limb amputations facing socket-related issues. However, understanding of its impact on musculoskeletal function remains limited.

Methods: This study compares pre- and 1-year post-osseointegration surgery outcomes, focusing on patient-reported measures and musculoskeletal system function during level-ground walking.

After traumatic brain injury (TBI), monocyte/macrophage infiltration is a key early step in the development of an inflammatory cascade that leads to substantial secondary damage. Intravenous (IV) immunomodulatory nanoparticle (IMP) administration after TBI limits inflammatory cell infiltration and reduces both behavioral decline and lesion size without any noticeable toxicity. Here we show that there is a dose-response relationship between the amount of IMP administered and tissue damage which plateaus at a well-tolerated dose.

Article Synopsis
  • Brain metastases (BrM) are a serious complication for cancer patients, occurring when cancer cells spread to the brain from other body parts, and there are limited effective treatments available.
  • Due to challenges in accessing patient samples, preclinical models are essential for studying how metastasis develops and responds to therapies, highlighting the need for reliable methods to measure metastatic burden.
  • This text describes a new semi-automatic machine-learning method that uses QuPath software to quickly and accurately quantify metastatic burden in mouse brain images while maintaining tissue integrity, making it more efficient and unbiased compared to traditional techniques.

Brain metastases (BrM) occur when malignant cells spread from a primary tumor located in other parts of the body to the brain. BrM is a deadly complication for cancer patients and currently lacks effective therapies. Due to the limited access to patient samples, preclinical models remain a valuable tool for studying metastasis development, progression, and response to therapy.
