Herein, we constructed a new type of hydrogel based artificial cells supporting long-lived protein synthesis, post-translational modification, and gene networks. We constructed the artificial cells by immobilizing the transcription and translation system from cytoplasmic extract onto the polyacrylamide hydrogel. With the continuous supply of energy and nutrition, the artificial cells were capable of stable protein expression for at least 30 days.

Plasmonic color generation has attracted much research interest because of the unique optical properties of plasmonic nanocrystals that are promising for chromatic applications, such as flat-panel displays, smart windows, and wearable devices. Low-cost, monodisperse plasmonic nanocrystals supporting strong localized surface plasmon resonances are favorable for the generation of plasmonic colors. However, many implementations so far have either a single static state or complexities in the particle alignment and switching mechanism for generating multiple displaying states.

We constructed the artificial cells by grafting anti-His-tag aptamer into the polymer backbone of the hydrogel particles, and then immobilizing the His-tagged proteinaceous factors of the transcription and translation system into the hydrogel particles. Long-term protein expression for at least 16 days was achieved by continuously flowing feeding buffer through the artificial cells.

We constructed the cell-mimic particles by immobilizing the proteinaceous factors of the cell-free transcription and translation system on the polymer backbone of hydrogel particles and encapsulating the plasmid template and ribosome inside the hydrogel. With the continuous supply of nutrients and energy, the protein expression in the cell-mimic particles remained stable for at least 11 days.
