STAT5 fulfills essential roles in hematopoietic stem cell (HSC) self-renewal and chronic myeloid leukemia (CML), a prototypical stem cell malignancy. However, the specific contributions of the two related genes STAT5A and STAT5B have not been determined. In this study, we used a RNAi-based strategy to establish participation of these genes to CML disease and persistence following targeted therapy.

Article Synopsis
  • Infection with HIV-1 and SIV in primates leads to significant issues in blood cell production, which are linked to problems in bone marrow function.
  • Research shows that during SIV infection in macaques, hematopoietic progenitor cells lose their ability to develop into multiple blood cell types due to decreased levels of proteins STAT5A and STAT5B; however, restoring STAT5B expression can reverse this loss.
  • The viral protein Nef plays a crucial role in this impairment and works by affecting the PPARgamma pathway, suggesting that drugs targeting PPARgamma could be potential treatments for AIDS and related blood disorders.
