Here, we present a previously undescribed approach to modify N-terminal sequences of recombinant proteins to increase their production yield in Escherichia coli. Prior research has demonstrated that the nucleotides immediately following the start codon can significantly influence protein expression. However, the impact of these sequences is construct-specific and is not universally applicable to all proteins.

The hippocampus (HPC) is essential for navigation and memory, tracking environmental continuity and change, including navigation relative to moving targets. CA1 ensembles expressing immediate-early gene (IEG) Arc and Homer1a RNA are contextually specific. While IEG expression correlates with HPC-dependent task demands, the effects of behavioral demands on IEG-expressing ensembles remain unclear.

Spatial games provide a simple and elegant mathematical model to study the evolution of cooperation in networks. In spatial games, individuals reside in vertices, adopt simple strategies, and interact with neighbors to receive a payoff. Depending on their own and neighbors' payoffs, individuals can change their strategy.

Article Synopsis
  • Genomic analysis can help assess risk and guide treatment for patients with large B cell lymphomas (LBCL) by identifying key genetic alterations linked to disease progression.
  • In a study of 698 newly diagnosed LBCL patients undergoing standard immunochemotherapy, MYC rearrangements and TP53 mutations were found to significantly increase the likelihood of disease progression within 24 months.
  • The research highlights the potential of using specific genomic features as biomarkers to tailor therapies for LBCL patients, which may improve treatment outcomes by addressing the unique characteristics of their disease.

Background: Large B cell lymphoma (LBCL) is the most common form of lymphoma. Polatuzumab vedotin (polatuzumab) is an effective therapy for patients diagnosed with LBCL; however, only limited information regarding pathologic features detected by clinical laboratory assays is available to determine which patients are most likely to benefit from polatuzumab based therapies.

Patients And Methods: We collected data from real world patients with relapsed or refractory LBCL whose tumors underwent next generation sequencing and were treated with polatuzumab based therapy at a single large academic cancer center.

The present paper deals with an analysis of total arsenic concentration using ICP-MS/MS and an analysis of concentration of several arsenic species, arsenite (As), arsenate (As), monomethylarsonate (MMA), dimethylarsenite (DMA), and trimethylarsine oxide (TMAO), using HPLC-ICP-MS/MS in the PM fraction of airborne urban aerosol. The samples were collected during two campaigns, in the autumn of 2022 and in the winter of 2023, at three locations within the central European city of Brno, with the aim to evaluate the seasonal and spatial variations in the PM composition. The results confirmed only the seasonal variability in the content of the methylated arsenic species in PM influenced by biomethylation processes.

Patients with B-cell lymphomas have altered cellular components of vaccine responses due to malignancy and therapy, and the optimal timing of vaccination relative to therapy remains unknown. Severe acute respiratory syndrome coronavirus 2 vaccines created an opportunity for new insights in vaccine timing because patients were challenged with a novel antigen across multiple phases of treatment. We studied serologic messenger RNA vaccine response in retrospective and prospective cohorts with lymphoma and chronic lymphocytic leukemia, paired with clinical and research immune parameters.

PPTC7 is a mitochondrial-localized phosphatase that suppresses BNIP3- and NIX-mediated mitophagy, but the mechanisms underlying this regulation remain ill-defined. Here, we demonstrate that loss of PPTC7 upregulates BNIP3 and NIX post-transcriptionally and independent of HIF-1α stabilization. Loss of PPTC7 prolongs the half-life of BNIP3 and NIX while blunting their accumulation in response to proteasomal inhibition, suggesting that PPTC7 promotes the ubiquitin-mediated turnover of BNIP3 and NIX.

Arsenic (As) levels in particulate matter (PM) are routinely monitored in cities of developed countries. Despite advances in the knowledge of its inorganic species in PM in urban areas, organic species are often overlooked with no information on their behaviour in urban parks - areas with increased potential for As biomethylation. Therefore, the aim of this study was to characterize As distribution, bioaccessibility, seasonal variation and speciation (As, As, MMA, DMA and TMAO) in PM-PM of an urban park.

Article Synopsis
  • - The study identifies the interaction between BTLA on T cells and HVEM on regulatory T cells as a major factor limiting the effectiveness of T cell-based immunotherapies in tumors.
  • - High levels of BTLA in CAR T cells are linked to poorer treatment outcomes, prompting researchers to delete BTLA to improve the T cells' tumor-fighting abilities.
  • - By removing BTLA, T cells show enhanced signaling and function, suggesting that targeting the BTLA-HVEM interaction could boost the success of CAR T cell therapies.

  • The study explores access to CART19 immunotherapy for large B-cell lymphomas among minority health populations (MHPs) compared to non-MHPs in different hospital settings.
  • It found that while MHPs had access to care for LBCL, their representation in CART19 therapy was lower, with only 6.7% at the Abramson Cancer Center and 4.2% at the Knight Cancer Institute receiving the treatment.
  • Outcomes for MHPs who did receive CART19 were similar to non-MHPs regarding response and survival, but the small sample size necessitates further research to confirm these findings.

Bendamustine has been retrospectively shown to be an effective and safe lymphodepletion regimen prior to the anti-CD19 chimeric antigen receptor T cell (CART) products tisagenlecleucel and axicabtagene ciloleucel, as well as the anti-BCMA CART products idecabtagene vicleucel and ciltacabtagene autoleucel. However, bendamustine as lymphodepletion prior to lisocabtagene maraleucel (liso-cel), a 4-1BB co-stimulated, fixed CD4:CD8 ratio anti-CD19 CART product, has not been described yet. Thus, we studied a cohort of sequentially-treated patients with large B-cell lymphomas who received bendamustine lymphodepletion before liso-cel at the University of Pennsylvania between 5/2021 and 12/2023 (n = 31).
