Gap junctions propagate toxic effects among tumor cells during chemotherapy, but could also enhance killing of normal cells by the same mechanism. We show that the effect of gap junctional intercellular communication (GJIC) on cisplatin toxicity differs between normal and tumor testicular cells. Downregulation of GJIC by each of several different manipulations (no cell contact, pharmacological inhibition, siRNA suppression) decreased cisplatin cytoxicity in tumor cells but enhanced it in normal cells.

Silica colloidal crystals were investigated for their potential as high surface area materials to enhance sensitivity over planar surfaces for microarrays using fluorescence detection. A relation was derived showing how crystal thickness and transmission, as well as colloid size, combine to determine the optically accessible surface area for enhancing sensitivity. Psychrobacter 273-4 was isolated from a 20,000-40,000-year-old Siberian permafrost core, which is characterized by low temperature, low water activity, and high salinity. To explore how 273-4 survives in the permafrost environment, proteins in four 273-4 samples cultured at 4 and 22 degrees C in media with and without 5% sodium chloride were profiled and comparatively studied using 2-D HPLC and MS. The method used herein involved fractionation via a pH gradient using chromatofocusing followed by nonporous silica (NPS) RP-HPLC and on-line electrospray mass mapping.

A method is developed to integrate a protein separation by monolithic capillary reversed-phase high-performance liquid chromatography to on-probe tryptic digestion for subsequent analyses by MALDI-TOF MS and MALDI-TOF/TOF MS. The method provides a means of directly interfacing separations to MALDI-MS, reducing the amount of time required for traditional procedures involving in-solution enzymatic digestion and sample cleanup prior to MALDI-MS analysis. When used with pI-based fractionation as a first dimension, it provides a means of analyzing complex mixtures of proteins with minimal sample handling and cleanup.

Crystals made from 200 nm silica colloids are hardened and chemically modified with chlorodimethyloctadecylsilane for use in electrically driven, reversed-phase separations. A van Deemter plot reveals extremely narrow peak widths for the separation of a cationic hydrophobic dye, DiI, with both the A and C terms 10-fold smaller than those for a conventional HPLC column. Electrically driven separations are demonstrated to be achieved in less than 10 s for three dyes differing in hydrophobicity and also for three peptides differing in electrophoretic mobility.

The hydrolysis reaction of L-alpha-distearoylphosphatidylcholine (DSPC) monolayers catalyzed by phospholipase A2 (PLA(2)) has been studied using polarization-modulated infrared reflection absorption spectroscopy (PM-IRRAS) with film balance measurements. The PM-IRRAS analysis provides quantitative information about the reaction efficiency at different surface pressures. It was found that the reaction efficiency of L-DSPC monolayer hydrolysis catalyzed by PLA(2) decreased with increasing surface pressure.

A multidimensional chromatographic method has been applied for the differential analysis of proteins from different strains of Escherichia coli bacteria. Background: Little is known about the influence of the site of research or publication on the impact of the research findings on clinical practice, particularly in developing countries. The International Clinical Epidemiology Network (INCLEN) is dedicated to improving the quality of health research in the Developing World through institutional capacity building for evidence based medicine, and provided the opportunity to examine the likely impact of research location and journal location on physicians' practice in a number of the participating countries.

Methods: Physicians from secondary and tertiary hospitals in six cities located in China, Thailand, India, Egypt and Kenya were enrolled in a cross-sectional questionnaire survey.
