PBAE-PEG based lipid nanoparticles for lung cell-specific gene delivery.

Delivery of modified mRNA encapsulated in lipid nanoparticles, exemplified by their successful use in COVID-19 vaccination, provides a framework for treating various genetic and acquired disorders. Herein, we developed PEGylated(PBAE-PEG) and non-PEGylated(PBAE) PBAE with lipids 4A3-SC8/DOPE/cholesterol/DOTAP to form lipid nanoparticles (LNPs) for mRNA delivery into different types of pulmonary cells in vivo. PBAE-PEG/LNP were highly active in transfecting HEK293T cells and air-liquid interfaced H441 cells in vitro.

Introduction: Emergency department overcrowding is a concern that predates the recent coronavirus disease pandemic. Overcrowding in the emergency department continues to worsen internationally. There are multiple combined strategies that help to maintain quality and safety by reducing patient wait times, left-without-being-seen rates, and the length of time a patient stays in the emergency department.

FOXA2 Cooperates with Mutant KRAS to Drive Invasive Mucinous Adenocarcinoma of the Lung.

Unlabelled: The endoderm-lineage transcription factor FOXA2 has been shown to inhibit lung tumorigenesis in in vitro and xenograft studies using lung cancer cell lines. However, FOXA2 expression in primary lung tumors does not correlate with an improved patient survival rate, and the functional role of FOXA2 in primary lung tumors remains elusive. To understand the role of FOXA2 in primary lung tumors in vivo, here, we conditionally induced the expression of FOXA2 along with either of the two major lung cancer oncogenes, EGFRL858R or KRASG12D, in the lung epithelium of transgenic mice.

Match-play movement demands of international and domestic women's rugby sevens players in an elite dual-level tournament.

Objectives: To characterize and compare match-play movement demands via Global Positioning Systems (GPS) between international and domestic women's rugby sevens players performing in a novel elite dual-level tournament, with consideration to position and tournament characteristics.

Design: Fifty-four rugby sevens players; twenty-one international(5 speed edges, 8 backs, 8 forwards), and thirty-three domestic(10 speed edges, 11 backs, 12 forwards) wore GPS devices during an elite dual-level tournament covering 2 seasons, with 367 full match-play data files analysed. Internationaland domestic players were distributed evenly between competing teams.

Trauma-Informed Pedagogy to Foster Resilience in Graduate Nursing Education.

The experiences of graduate nursing students during the COVID-19 pandemic necessitate a trauma-informed approach to education. Three hundred graduate nursing students responded to a discussion assignment in a doctoral-level health care policy course. Forty-two units (GPSports EVO; 10 Hz, n = 13: GPSports HPU; 5 Hz, n = 14: and Catapult S5; 10 Hz, n = 15) were investigated across 3 experiments: a 40-m linear track with all units pushed on a trolley, a sport simulation circuit with all units pulled on a sled, and a similar circuit with 3 models of units placed in a modified GPS vest worn by an athlete between the scapulae.

Evidence-Based Facebook Recruitment of Study Participants.

Traditional methods for research study recruitment such as snail mail lists and posting flyers may fail to reach the tech-savvy participants needed for today's healthcare studies. Word of mouth can be effective for recruiting a few participants but can rarely accomplish the numbers needed for a representative sample. Social media can be a viable avenue to reach increased numbers of sample participants; however, a good understanding of the risks and benefits of using social media is needed before embarking on active recruitment.

Simulation-Based Training Program to Improve Cardiopulmonary Resuscitation and Teamwork Skills for the Urgent Care Clinic Staff.

Introduction: Lack of high-quality cardiopulmonary resuscitation (CPR) and effective team communication during cardiovascular emergencies could lead to poor patient outcomes and adverse events. Studies have shown that CPR psychomotor skills declined 3 months and plateaued up to 6 months after the completion of CPR training. The Urgent Care Clinic (UCC) interprofessional staff did not receive consistent training on CPR and teamwork skills beyond what was received from the biannual American Red Cross (ARC) Basic Life Support (BLS) training and semiannual Resuscitative Care Function mock cardiac arrest.

CRISPRi-mediated functional analysis of NKX2-1-binding sites in the lung.

The transcription factor NKX2-1/TTF-1 is involved in lung pathophysiology, including breathing, innate defense and tumorigenesis. To understand the mechanism by which NKX2-1 regulates genes involved in such pathophysiology, we have previously performed ChIP-seq and identified genome-wide NKX2-1-binding sites, which revealed that NKX2-1 binds to not only proximal promoter regions but also multiple intra- and inter-genic regions of the genes regulated by NKX2-1. However, the roles of such regions, especially non-proximal ones, bound by NKX2-1 have not yet been determined.

Increasing Outpatient Clinic Healthcare Provider Engagement Through Nursing Leader Communication.

Maintaining engaged nurses and other health care providers were challenging at a multi-specialty outpatient clinic where engagement scores were trending downward. This quality improvement (QI) was developed to improve engagement by implementing structured, evidence-based practice (EBP) interventions to increase nursing leader communication, visibility, and feedback. The Utrecht Work Engagement Scale (UWES-17) compared pre-and post-intervention effectiveness from week one to week eight.

Implementing a Mentorship Program for New Nurses During a Pandemic.

This article describes the implementation of an evidence-based mentoring program for new registered nurses (RNs) hired into medical-surgical units in a small community-based hospital during the unfolding of the SARS-Cov2 (COVID-19) pandemic. The hospital's nursing leadership supported the program implementation during the COVID-19 pandemic to provide a broader support system to new RNs to improve nurse retention. During a response to the pandemic, the medical-surgical units faced numerous process changes in a short time, which further reinforced the urgency of an additional support system for the newly hired RNs.

Background: Little is known about the extent to which routine care management of peripheral arterial disease (PAD) and intermittent claudication (IC) align with best practice recommendations on exercise therapy. We conducted a scoping review to examine the published literature on the availability and workings of exercise therapy in the routine management of patients with PAD and IC, and the attitude and practice of health professionals and patients.

Methods: A systematic search was conducted in February 2018.

Patient perception of caregiver empathy has been studied internationally for decades. Although leadership empathy has been described as having a critical influence on staff retention, this subject is less evident in the literature. This article described a study undertaken to gain a baseline understanding of nurses' perceptions of leader empathy from those they identify as their managers.

Efficacy and Feasibility of Pain management and Patient Education for Physical Activity in Intermittent claudication (PrEPAID): protocol for a randomised controlled trial.

Background: Physical activity (PA) improves functional capacity and quality of life and provides secondary prevention benefits in individuals with peripheral arterial disease (PAD) and intermittent claudication (IC). However, pain and patient lack of knowledge are key barriers to the uptake of, and adherence to, PA recommendations. This trial will test the efficacy and feasibility of a non-invasive pain management intervention with and without patient education to improve PA in individuals with PAD and IC.

The Influence of Socio-economic Deprivation on Mobility, Participation, and Quality of Life Following Major Lower Extremity Amputation in the West of Scotland.

Objective: Lower extremity amputation (LEA) is more common in people from lower socio-economic groups. This study examined this further by investigating the influence of socio-economic status on mobility, participation, and quality of life (QoL) after LEA.

Methods: Prospective data were gathered for all LEAs performed in one year in one Scottish Health Board, commencing March 2014.

Peroneal artery entrapment syndrome (PRAES): a rare cause of ischaemic toes.

A 75-year-old man presenting with intermittent discolouration of his left toes was referred to vascular surgery with suspicion of embolic vascular disease. A contrast-enhanced MR angiogram was performed which revealed bilateral dominant peroneal arteries (PRAs). There was evidence of short atherosclerotic stenosis directly at the point where the left PRA passes through the tibiofibular interosseous membrane which we postulate to be the source of the emboli.

The hit-and-return system enables efficient time-resolved serial synchrotron crystallography.

We present a 'hit-and-return' (HARE) method for time-resolved serial synchrotron crystallography with time resolution from milliseconds to seconds or longer. Timing delays are set mechanically, using the regular pattern in fixed-target crystallography chips and a translation stage system. Optical pump-probe experiments to capture intermediate structures of fluoroacetate dehalogenase binding to its ligand demonstrated that data can be collected at short (30 ms), medium (752 ms) and long (2,052 ms) intervals.
