Article Synopsis
  • The study investigates the effects of ethanol on skeletal muscle protein balance in alcohol-associated cirrhosis, focusing on how it disrupts signaling pathways like mTORC1 and AMPK.
  • Ethanol leads to increased muscle autophagy by dephosphorylating mTORC1 while inhibiting AMPK, suggesting a complex interaction in energy regulation and muscle maintenance, termed a "pseudofed" state.
  • The research identifies protein phosphatase 2A as a significant player in these processes and shows that altering the activity of PI3Kγ can help counteract the negative effects of ethanol on muscle signaling and function.

Neuropilin-2, a cell surface receptor involved in angiogenesis and axonal guidance, has recently been shown to be a critical mediator of tumor-associated lymphangiogenesis. Given that lymphangiogenesis is a major conduit of metastasis in melanomas and that blocking neuropilin-2 function in vivo is effective in inhibiting tumor cell metastasis, we sought to determine the clinical relevance of neuropilin-2 expression in cutaneous melanoma. Immunohistochemical analysis of neuropilin-2 expression was evaluated in nevomelanocytic proliferations that included a tissue microarray and histologic sections from samples of primary melanomas (n = 42; 40 for tissue microarray, 2 for histologic sections), metastatic melanomas (n = 30; 22 for tissue microarray, 8 for histologic sections), and nevi (n = 30; 5 for tissue microarray, 25 for histologic sections), as well as a panel of normal human tissues and select nonmelanocytic tumors.

Tumor-endothelium interactions are critical for tumor survival and metastasis. Melanomas can rapidly metastasize early in tumor progression, but the dependence of this aggressive behavior on tumor-stromal interaction is poorly understood. To probe the mechanisms involved, we developed a heterotypic coculture methodology, allowing simultaneous tracking of genomic and phenotypic changes in interacting tumor and endothelial cells in vitro.

Since its inception, minimally invasive surgery has made a dramatic impact on all branches of surgery. Video-assisted thoracic surgery (VATS) lobectomy for early-stage non-small cell lung cancer (NSCLC) was first described in the early 1990s and has since become popular in a number of tertiary referral centers. Proponents of this relatively new procedure cite a number of potentially favorable perioperative outcomes, possibly due to reduced surgical trauma and stress.

A microfluidic electroporation technique is demonstrated based on the operation of an elastomeric valve in a poly(dimethylsiloxane) (PDMS) fabricated microchip and a common dc power supply. The pulse needed for permeabilization of the cell membrane is generated by temporarily interrupting the circuit using the valve. The electropermeabilization of suspended and adherent Chinese hamster ovary cells with green DNA dye SYTOX is demonstrated.

We sought to determine the distribution of resistance and the tetracycline resistance genes among bacteria isolated from a swine confined animal feeding facility where tetracycline-containing feed had been in use for over 20 years. Samples collected from feed, hogs, hog houses, waste lagoon, soil, surface water and well water were screened for the presence of (a) resistant Escherichia coli and enterococci and (b) tetracycline-resistant strains of all species. Genomic DNA was extracted from the latter strain collection and fragments from 16S rDNA and ten tetracycline resistance genes (tetA, tetB, tetC, tetE, tetH, tetL, tetM, tetS, tetT and rumB) were polymerase chain reaction-amplified and a partial nucleotide sequence was obtained.

Recent studies with adolescent rodents offer valuable information regarding the neurochemical and behavioral effects of adolescent nicotine exposure. One hundred twenty-one male and 125 female adolescent (35 days of age) C57BL/6J mice were tested for voluntary nicotine consumption by providing 24-h access to both saccharin-only (SAC) and one of six nicotine-containing solutions [10, 25, 50, 75, 100, 200 ug (-)-freebase nicotine/ml in 2% SAC] in the home cage for 7 days. Although males and females drank similar volumes (ml) of nicotine, the female mice consumed more nicotine adjusted for body weight (mg/kg) and as a percentage of total fluid intake than did the male mice.

Time estimation is used as an index of attention processes and may be sensitive to self-administration and withdrawal of psychoactive drugs such as nicotine, the primary addictive ingredient in tobacco. To test this hypothesis, 22 nonsmokers (12 male, 10 female) and 20 daily cigarette smokers (12 male, 8 female) were asked to estimate the duration of a 45-second period of time in a laboratory setting. Smokers participated in two sessions: once after smoking ad-lib and once after objectively confirmed 24-hour smoking abstinence.

The grass stage is an inherent and distinctive developmental trait of longleaf pine (Pinus palustris), in which height growth in the first few years after germination is suppressed. In operational forestry practice the grass stage extends for two to several years and often plays a role in planting failures and decisions to plant alternative species. Interspecies hybrids involving loblolly (P.

Maternal cigarette smoking is a risk factor for adolescent smoking. One possible explanation for increased smoking by human adolescents after maternal nicotine exposure is that exposure increases nicotine preference. However, it is difficult to separate the biological and social causes of smoking behavior in humans.

The present study examined the relationship between regular caffeine consumption and time estimation. Sixty participants (aged 18-57 years, mean 24 years) completed a 47 s time estimation exercise and questionnaires related to daily caffeine consumption and perceptions of time. We hypothesized that the effects of caffeine on time estimation would follow a U-shaped pattern such that individuals who reported moderate amounts of daily caffeine would be more accurate in their perceptions of time than would those who reported high amounts of caffeine intake or no daily caffeine consumption.

The purpose of this study was to examine in a systematic, controlled fashion the reactions of smokers to scientifically correct information about the risks of smoking Light cigarettes (about 6-15 mg tar by the FTC method). Random-digit dialing, computer-assisted telephone interviews were used to locate daily smokers of Light cigarettes. In an experimental design, smokers were randomly assigned to listen (n = 293) or not (n = 275) to a persuasive simulated radio message on the risks of Light cigarettes; 108 of those who did not listen to the message in the first part of the interview were played the message in the second part, to evaluate some repeated-measures effects.

The Tower of Hanoi and Tower of London have become well-established executive function tasks that presumably tap cognitive skills mediated by the frontal cortex. It has been assumed that the two tower tasks are more or less interchangeable and that both measure working memory and inhibition processes. These assumptions were tested in a study involving 37 normal college volunteers (M age = 20 years).

Individual bees often prefer flowers of the same species that they are already foraging on, and other individual bees prefer other flowers. This "floral constancy" has classically been explained as a learned behavior by which bees avoid wasting time switching between handling techniques. Choice trails were given to Bombus vagans workers that were freely foraging in mixed and pure fields of Trifolium pratense, T.

Three major themes related to the improvement of salt tolerance in forest tree species are examined. First, evidence demonstrating that substantial intraspecific variation in salt tolerance exists in many species is presented. This evidence is important because it suggests that efforts to improve salt tolerance through conventional plant breeding techniques are justified.

The immunization compliance among 2-year-olds in a prepaid healthcare system in Indianapolis was studied. Since appointments and immunizations are covered benefits, immunization rates should approach 100%; complete immunizations were found in only 72%. Problems identified included transient membership, failure to determine immunization status on patients, reluctance to administer immunizations to children with mild illnesses, asking patients to return for administration of immunizations without a physician exam and record-keeping errors.

Premature babies can, and should, breastfeed. Although establishing a milk supply and the transition to breastfeeds can be difficult, both are possible. Because early bottle-feeds can impede later attempts to breastfeed, the newborn intensive care unit at Methodist Hospital in Indianapolis developed a protocol for successfully breastfeeding premature infants that excludes bottle-feeding.

The etiology of necrotizing enterocolitis (NEC) is unknown. We conducted a retrospective, case-controlled study of 23 infants hospitalized during a five-year period who developed NEC after the 11th day of life (late-onset NEC). We evaluated 58 different factors among the case and control populations to determine if there were any statistically significant differences between the two populations.
