A disturbance in the balance between cell proliferation and cell loss, or apoptosis, may underlie neoplastic development. Therefore, we determined spontaneous apoptotic and proliferative rates in normal, hyperplastic, adenomatous, and malignant colorectal epithelia. In paired sections, DNA strand breaks were detected using the terminal deoxynucleotidyltransferase-mediated dUTP nick-end labeling assay, and apoptotic cells were also identified in H&E-stained slides by morphological criteria.

For up to 8 h after irradiation, levels of apoptosis were higher in the C57BL/6J strain for all doses assayed. The heritability of the strain difference in the extent of apoptosis resulting from irradiation was investigated using a breeding study.

Levels of radiation-induced jejunal crypt cell apoptosis were compared in C57BL/6J, C3Hf/Kam and C3H/HeJ mice. Apoptosis levels were consistently lower in the C3H strains than in C57BL/6J. Although other explanations are possible, the strain difference is most likely to have a genetic basis, and in fact a preliminary analysis of the F2 progeny of C3H/HeJ and C57BL/6J mice indicates that more than one gene is involved.

Expression of a beta2m transgene controlled by the rat insulin promoter (RIP-beta2m) in NODbeta2mnull mice resulted in reconstitution of IFN-gamma-inducible cell surface MHC class I protein on pancreatic beta-cells. These mice developed insulitis, but did not develop diabetes.

In normothermic rats, TNF alone (10(5) or 10(6) U/kg) did not cause hypotension, but increased MAP (p < 0.05).

Quantification of FITC-phalloidin binding by fluorescence-activated cell sorting reveals that both ATP and insulin stimulate actin polymerization with distinctive kinetics in U937 cells. The response to ATP is rapid and dose-dependent with an EC50 of 200 nM, and is abolished by pre-incubation with the Ca2+ chelator BAPTA-AM.
View Article and Find Full Text PDFPeptides carrying an immunodominant T-helper cell epitope delineated from the rabies virus nucleoprotein either alone or in combination with a linear B-cell epitope from the same protein were incorporated into three different formulations of poly(DL-lactide-co-glycolide) (PLG) which were distinct in their composition, and consequently in their peptide release rates. In vitro peptides incorporated into any of the PLG formulations stimulated a peptide-specific T-cell line. Upon subcutaneous immunization of mice, the PLG formulation that showed the fastest peptide release rate induced the best immune response.

We assessed the effects of 41 potential chemopreventive agents in the F344 rat using the inhibition of carcinogen-induced aberrant crypt foci (ACF) in the colon as the measure of efficacy. ACF were induced by the carcinogen azoxymethane in F344 rats by two sequential weekly injections at a dose of 15 mg/kg. Two weeks after the last azoxymethane injection, animals were evaluated for the number of aberrant crypts detected in methylene blue-stained whole mounts of rat colon.

The occurrence of spontaneous apoptosis was different when comparing primary to metastatic tumor growth. In the primary MTLn3 tumor growing at the mammary fat pad inoculation site we observed an inverse association between tumor growth and apoptosis.

Purpose: To determine the relationship between pretreatment apoptosis levels and clinical-to-pathologic downstaging resulting from preoperative radiotherapy.
Methods And Materials: Between 1960-1983, 338 patients were dispositioned to receive preoperative radiotherapy 4-6 weeks prior to radical cystectomy for muscle-invasive transitional cell carcinoma of the bladder. Of these, adequate hematoxylin and eosin stained tissue sections for morphologic analysis of apoptosis were available in 158 patients.
Preview Clostridial myonecrosis (gas gangrene) is an uncommon sequela of traumatic injury. Because the infection can become rapidly fatal, early recognition and expeditious treatment are critical. In addition to wide surgical debridement and treatment with broad-spectrum antibiotics, hyperbaric oxygen therapy has been shown to have synergistic benefit.

We have conducted a series of studies using murine tumor models and found that the ability of the tumor cells to undergo apoptosis correlates with tumor response to radiation. The best correlation was with the pretreatment apoptotic index, suggesting that apoptosis in some tumors may govern radiocurability by regulating the number of tumor clonogens.

Deficiencies in circulating anticoagulant protein C (PC) occur in patients undergoing hematopoietic stem cell transplantation. These deficiencies may predispose to thrombotic and other complications, and may contribute to the morbidity of transplantation. In most patients, PC reaches a nadir 14 days after the preparative regimen and then begins to increase toward normal.

We have purified two forms of phosphoinositide 3-kinase (PI3K) that are activated by heterotrimeric G-protein beta gamma-subunits. These novel isoforms of PI3K are structurally distinct to those forms of PI3K which have already been cloned. They are both heterodimers made up of a p120 and a p101 and a p117 and a p101 protein.

The p53 gene is frequently altered in human cancers, including breast cancer, and such alterations are thought to result in genomic instability and aneuploidy, 2 hallmarks of anaplastic cells. GUS activity in Agrobacterium 8999 was reduced to levels in negative controls of LBA4404. In vitro and in vivo activities of this catheter were compared with those of an aseptic catheter coated with chlorhexidine and silver sulfadiazine (CH/SS). The SIC demonstrated broad-spectrum in vitro inhibitory activity against bacteria and Candida albicans comparable to that of the CH/SS-coated catheter.
