DNA methylation of the TPMT gene and azathioprine pharmacokinetics in children with very early onset inflammatory bowel disease.

Background: Thiopurine methyltransferase (TPMT) is a crucial enzyme for azathioprine biotransformation and its activity is higher in very early onset inflammatory bowel disease (VEO-IBD) patients than in adolescents with IBD (aIBD).

Aims: The aims of this pharmacoepigenetic study were to evaluate differences in peripheral blood DNA methylation of the TPMT gene and in azathioprine pharmacokinetics in patients with VEO-IBD compared to aIBD.

Methods: The association of age with whole genome DNA methylation profile was evaluated in a pilot group of patients and confirmed by a meta-analysis on 3 cohorts of patients available on the public functional genomics data repository.

Overcome the limits of multi-contaminated industrial soils bioremediation: Insights from a multi-disciplinary study.

Phytoremediation can be a promising and sustainable strategy to recovery Potentially Toxic Elements (PTEs) contaminated soils. However, at the field level, this tool can be limited by many issues. Herein, we combined native plant species with different cover type (mono and poly culture) in an in-field mesocosm experiment for the remediation of multi-contaminated soils from Bagnoli brownfield site (Southern Italy).

Investigation and Assessment for an effective approach to the reclamation of Polycyclic Aromatic Hydrocarbon (PAHs) contaminated site: SIN Bagnoli, Italy.

Native plant species were screened for their remediation potential for the removal of Polycyclic Aromatic Hydrocarbons (PAHs) contaminated soil of Bagnoli brownfield site (Southern Italy). Soils at this site contain all of the PAHs congeners at concentration levels well above the contamination threshold limits established by Italian environmental legislation for residential/recreational land use, which represent the remediation target. The concentration of 13 High Molecular Weight Polycyclic Aromatic Hydrocarbons in soil rhizosphere, plants roots and plants leaves was assessed in order to evaluate native plants suitability for a gentle remediation of the study area.

Identification of native-metal tolerant plant species in situ: Environmental implications and functional traits.

A study was undertaken to identify suitable native plants for the phytoremediation of the second largest integrated steelworks in Italy (Bagnoli brownfield site). A phytoecological survey allowed us to identify 139 plant taxa belonging to 58 different families. The most represented families were in the decreasing order Poaceae > Fabaceae > Asteraceae > Apiaceae.

Novel NOD2 Mutation in Early-Onset Inflammatory Bowel Phenotype.

Background: Nucleotide-binding oligomerization domain 2 (NOD2) is a key intracellular protein of the innate immune system. NOD2 variants are associated with inflammatory bowel disease (IBD) and other inflammatory phenotypes. We described the case of a baby with a very early-onset IBD who is characterized by a rare homozygous variant in NOD2, found through whole-exome sequencing, Its pathogenic effect was investigated through bioinformatics and functional studies.

Community-acquired Clostridium difficile infection in children: A retrospective study.

Background: Community acquired-Clostridium difficile infection (CDI) has increased also in children in the last years.

Aims: To determine the incidence of community-acquired CDI and to understand whether Clostridium difficile could be considered a symptom-triggering pathogen in infants.

Methods: A five-year retrospective analysis (January 2007-December 2011) of faecal specimens from 124 children hospitalized in the Niguarda Ca' Granda Hospital for prolonged or muco-haemorrhagic diarrhoea was carried out.

Growth hormone deficiency, anorectal agenesis, and brachycamptodactyly: a phenotype overlapping Stratton-Parker syndrome.

Stratton and Parker [1989] described a 17-month-old boy with the previously unreported combination of growth hormone (GH) deficiency, Wormian bones, mild developmental delay, brachycamptodactyly, heart defects, kidney hypoplasia, imperforate anus, bilateral cryptorchidism, and facial anomalies. A similar case was later reported by Gabrielli et al. [1994], who suggested the existence of a "Stratton-Parker syndrome.

Growth hormone secretory pattern and response to treatment in children with short stature followed to adult height.

Objective: To compare the relative utility of GH stimulation tests and assays of spontaneous GH secretion as predictors of change in height standard deviation score at the end of GH treatment in children with short stature.

Patients And Methods: We retrospectively studied 116 children (67 boys and 49 girls) with subnormal growth rates and short stature, defined as a height of more than 2SD below the mean for age and sex. The patients were classified according to their pattern of findings on baseline pharmacological GH stimulation tests and a 12-h assay of nocturnal spontaneous GH secretion.

Determination of As in environmental solid matrix.

In line with recent European environmental guidelines on biomasses, one of the most important parameters to take into account is the As concentration, especially when present in biomasses and complex matrices. The goal of the present study is to give information about possible technical-analytical problems during the determinations of such elements by means of different instrumental spectroscopy techniques, in particular inductively coupled plasma atomic emission (ICP-AES) and atomic absorption (AAS), using two different wavelengths, 188.98 nm and 193.

Arsenic and heavy metals in leachate from a real landfill and a laboratory landfill.

A preliminary study, based particularly on Arsenic and some heavy metals determination (Cd, Cr, Hg and Pb) in three leachate tipologies produced from municipal waste landfill plant, lab-scale aerobic landfill, lab-scale anerobic landfill was undertaken. Leachate was collected and analysed monthly; samples were homogenized and then, all heavy metals were determined after mineralization, except for Hg, determined before mineralization, due to his high volatility. In all samples pH was measured.
