Experiments done with a temperature"sensitive mutant of herpes simplex virus type 1 (HSV-1) have revealed that one of the virisn glycoproteins, designated VP7(B2), is apparently not required for the production of enveloped virus particles, whereas it does play a critical role in virion infectivity. The mutant, designated HSV-1[HFEM]tsB5, fails to accumulate VP7(B2) at nonpermissive temperature and produces virions that lack detectable quantities of this glycoprotein and that have very low specific infectivity. The poor infectivity of the virions is most readily explained by failure of penetration into the host cell rather than by failure of adsorption to cells because it was shown that the VP7(B2)-deficient virions can bind to cells and that polyethylene glycol, an agent known to promote membrane fusion, can significantly enhance infectivity of the adsorbed virions.

The virions of different herpesviruses are similar with respect to the number and kinds of constituent polypeptides, in spite of variability in the structures of individual polypeptides. The total number of virion polypeptides and glycopeptides ranges from 20 to 30 for different viruses and, in general, no more than one-quarter of these polypeptides is detectable in naked nucleocapsids, implying that most of the virion polypeptides are acquired during the process of envelopment. Although the functions of most individual structural proteins have not been identified, one can predict that the nucleocapsid proteins serve primarily structural roles or may mediate packaging of the viral genome, that the non-glycosylated envelope proteins play essential roles in the process of envelopment and that the glycoproteins, which are probably all exposed to the virion surface, mediate adsorption to and penetration of the host cell.

Some of the factors that regulate membrane fusion resulting in polykaryocyte formationhave been investigated, using the model system of human cells infected with mutants of herpes simplex virus (HSV). One of the mutant viruses used in this study (MP) failed to produce the viral glycoprotein designated C2--a nonlethal defect that has previously been correlated with the polykaryocyte-inducing phenotype of this and other mutant strains (wild-type strains of HSV usually induce the aggregation of infected cells rather than their fusion). The other mutant virus (tsB5), a temperature-sensitive conditional-lethal mutant, failed to produce glycoprotein B2 at non-permissive temperature, whereas the synthesis of all other viral products appeared to be normal.

The cell line used in this study is a derivative of tk-deficient mouse L cells that, after exposure to UV-inactivated HSV-1, had acquired the HSV-1 gene for tk (which we term a resident viral gene) and consequently expressed the tk+ phenotype (LVtk+ cells). Productive infection of these cells with HSV-1(tk-) at appropriate multiplicities caused significant enhancement of the viral tk activity.

In Experiment 1, rats with serial unilateral (SU), serial bilateral (SB), or one-stage bilateral (B) visual cortex lesions (areas 17, 18 and 18a) were compared in their retention of a preoperatively learned brightness discrimination. Both the first-stage and second-stage lesions for the SU and SB groups involved the same amount of cortical tissue and the same composite areas of visual field projection in primary and extraprimary visual cortex. Groups SU and SB showed substantial savings of the brightness discrimination after the completed bilateral visual cortex removals, while Group B showed a complete postoperative loss.

The results demonstrate that the major glycosylated proteins detected in infected cells are derived from four antigenically distinct polypeptides. Three of these polypeptides become glycosylated in two discrete stages, yielding partially glycosylated intermediates and fully glycosylated products.

The visual receptive fields of 213 cells in the lateral suprasylvian visual cortex (LS, or Clare-Bishop area) were studied in cats anesthetized with nitrous oxide. Eighty-one percent of the cells were directionally selective. Six monocularly deprived (MD) and four normal cats were trained monocularly on two-choice form and pattern discriminations. MD cats trained through the initially deprived eye were able to learn the discriminations; however, they required many more trials than normals. Retention tests showed that MD cats have nearly perfect retention of the discriminations over periods of up to 4 months.
