  • Ovarian tumor domain-containing protease 1 (OTUD1) is crucial for regulating immune responses and plays a significant role in the development of sepsis, as demonstrated in multiomics analyses of clinical and experimental models.
  • The study found that increased OTUD1 expression negatively affects inflammatory responses and helps protect against sepsis-induced lung damage by inhibiting specific signaling pathways, namely MAPK and NF-κB.
  • Additionally, it was revealed that OTUD1 can deubiquitinate the protein TIPE2, which also provides protective effects against sepsis-related injuries, suggesting that OTUD1 could be an important therapeutic target for treating lung injuries associated with sepsis.

Sepsis-induced acute lung injury has been deemed to be an life-threatening pulmonary dysfunction caused by a dysregulated host response to infection. The modification of N6-Methyladenosine (m6A) is implicated in several biological processes, including mitochondrial transcription and ferroptosis. Ferroptosis is an iron-dependent type of programed cell death, which plays a role in sepsis-induced acute lung injury (ALI).

Article Synopsis
  • Researchers are studying how a process called autophagy, which helps cells clean up their waste, is affected during sepsis (a serious infection).
  • They found that when a protein called Erbin is missing, it makes the inflammatory response worse and can cause more damage to organs during sepsis.
  • The study suggests that Erbin helps protect against this damage by fixing problems in the autophagy process and working with other proteins to help cells respond better to infection.

Background: This study aims to investigate the correlation between Erbin and sepsis, and the role of Erbin on the pyroptosis pathway in acute kidney injury caused by sepsis and NLRP3/caspase-1/Gasdermin D pathway.

Methods: In the study, lipopolysaccharide (LPS) treatment or cecal ligation and puncture (CLP) surgery on mice were used to stimulate the in vitro and in vivo sepsis-induced renal injury model. The male C57BL/6 of wild-type mice (WT) and Erbin-knockout mice (Erbin, EKO) were randomly divided into four groups (WT + Sham, WT + CLP, EKO + Sham, EKO + CLP).
