Article Synopsis
  • Researchers have developed the fungus Aspergillus oryzae to produce a llama-derived antibody fragment (V(HH)) that targets the epidermal growth factor receptor (EGFR) effectively.
  • By integrating a Taka-amylase A signal sequence and a specific amino acid segment from Rhizopus oryzae lipase, they significantly increased the yield of this antibody fragment to 73.8 mg/1.
  • The resulting V(HH) fragments can be released from their fusion tags during cultivation and demonstrate strong binding affinity to EGFR, making them immunologically active.

A novel cell surface display system in Aspergillus oryzae was established by using a chitin-binding module (CBM) from Saccharomyces cerevisiae as an anchor protein. CBM was fused to the N or C terminus of green fluorescent protein (GFP) and the fusion proteins (GFP-CBM and CBM-GFP) were expressed using A. oryzae as a host.
