Depletion of TP53 in Human Pluripotent Stem Cells Triggers Malignant-Like Behavior.

Human pluripotent stem cells (hPSCs) tend to acquire genetic aberrations upon culture in vitro. Common aberrations are mutations in the tumor suppressor TP53, suspected to confer a growth-advantage to the mutant cells. However, their full impact in the development of malignant features and safety of hPSCs for downstream applications is yet to be elucidated.

Grazing and topography control nutrient pools in low Arctic soils of Southwest Greenland.

Unlabelled: Soil nutrient pools in the dry low Arctic are likely to be released under climatic change and this bioavailability has the potential to increase both terrestrial and aquatic productions. As well as the direct effect of warming, external disturbances such as nutrient deposition and grazing can also drive ecosystem change. This study in the low Arctic Kangerlussuaq area of southwest Greenland compared soil nutrient pools in terms of both topographic position on a catena and by soil depth in two small catchments with contrasting muskox abundance.

Refining lunar impact chronology through high spatial resolution (40)Ar/(39)Ar dating of impact melts.

Quantitative constraints on the ages of melt-forming impact events on the Moon are based primarily on isotope geochronology of returned samples. However, interpreting the results of such studies can often be difficult because the provenance region of any sample returned from the lunar surface may have experienced multiple impact events over the course of billions of years of bombardment. We illustrate this problem with new laser microprobe (40)Ar/(39)Ar data for two Apollo 17 impact melt breccias.

Episodic entrainment of deep primordial mantle material into ocean island basalts.

Chemical differences between mid-ocean ridge basalts (MORBs) and ocean island basalts (OIBs) provide critical evidence that the Earth's mantle is compositionally heterogeneous. MORBs generally exhibit a relatively low and narrow range of (3)He/(4)He ratios on a global scale, whereas OIBs display larger variability in both time and space. The primordial origin of (3)He in OIBs has motivated hypotheses that high (3)He/(4)He ratios are the product of mantle plumes sampling chemically distinct material, but do not account for lower MORB-like (3)He/(4)He ratios in OIBs, nor their observed spatial and temporal variability.

The usability of a modified Astrand bicycle test to assess the aerobic capacity in patients with musculoskeletal pain and healthy controls.

Purpose: The aim of this study was to assess the usability of a modified Astrand bicycle test in subjects with musculoskeletal pain disorder (MSPD) and healthy controls.

Methods: In a random cross-over design, 31 subjects with a MSPD who have been referred to rehabilitation medicine and 33 healthy subjects matched for age, gender, height and body weight, performed an Astrand bicycle test and a modified test with a time interval of 7 days. Drop-out rates, VO2max values (in mL/kg lean body mass (LBM) min(-1)), intraclass correlation coefficients (ICCs) and critical difference (Bland-Altman plots) for both tests within each group of subjects were compared.

[Prevention and treatment of somatic complications arising from the use of antipsychotics].

Background: Antipsychotics are effective drugs that are prescribed frequently for a large group of patients. However, they also have many side-effects which can lead ultimately to serious somatic complications. These complications fall into various categories: metabolic, cardiovascular, neurobiological, haematological, gastro-intestinal and urogenital.

Flow of mantle fluids through the ductile lower crust: helium isotope trends.

Heat and mass are injected into the shallow crust when mantle fluids are able to flow through the ductile lower crust. Minimum 3He/4He ratios in surface fluids from the northern Basin and Range Province, western North America, increase systematically from low crustal values in the east to high mantle values in the west, a regional trend that correlates with the rates of active crustal deformation. The highest ratios occur where the extension and shear strain rates are greatest.

Clinical application of the polymerase chain reaction in the detection of HLA-B27 alleles.

Most patients with ankylosing spondylitis (AS) express HLA-B27 antigens, which in routine clinical practice are conventionally detected by serological assays. Recent reports have been published demonstrating that HLA-B27 can also be detected by the polymerase chain reaction (PCR), with the purported advantages that the latter is faster, cheaper and more reliable than serological detection. In this study the technical reliability of HLA-B27 detection by PCR was demonstrated by the successful amplification of all the defined types of HLA-B27 alleles, while relevant non-HLA-B27 alleles could not be amplified.

Hemodynamic parameters for venous valve incompetence in the lower extremity.

Doppler and photoplethysmography studies were compared with invasive venous pressure measurements in ten healthy volunteers and 20 patients with a postthrombotic syndrome to assess the accuracy of noninvasive studies in detecting venous valve incompetence in the lower extremity. Using a refilling time of 20 seconds or less for valve incompetence, photoplethysmography was as accurate as invasive pressure measurements. In the Doppler studies there were two false-positive and three false-negative studies as compared with the invasive method.

Hemodynamic evaluation of transluminal iliac artery balloon dilatation.

In order to document the hemodynamic results of transluminal iliac artery balloon dilatation, 23 aortoiliac segments were evaluated before and after treatment. Hemodynamic parameters were: intra-arterial common femoral pressure measurements, indirect ankle pressure measurements and femoral velocity waveform analysis. The segments were divided into group (a) aortoiliac segments with an open superficial femoral artery (n = 8), and group (b) aortoiliac segments with an occluded superficial femoral artery (n = 15).

The clinical value of preoperative indirect systolic ankle pressure measurements in wound healing after amputation of the lower extremity.

With the intention towards more distal amputation of the lower extremity for peripheral vascular disease, failure of amputation wound healing remains a common clinical problem. Because calf blood flow correlates well with the indirect systolic ankle pressure, this pressure could be a prognostic guide to the outcome of amputation wound healing. To evaluate the clinical role of systolic ankle pressure measurements for selecting the most appropriate level of amputation, data of 93 patients undergoing 100 amputations were studied retrospectively.
