Topological and deletion analysis of CorS, a Pseudomonas syringae sensor kinase.

A modified two-component regulatory system consisting of two response regulators, CorR and CorP, and the histidine protein kinase CorS, regulates the thermoresponsive production of the phytotoxin coronatine (COR) in Pseudomonas syringae PG4180. COR is produced at the virulence-promoting temperature of 18 degrees C, but not at 28 degrees C, the optimal growth temperature of PG4180. Assuming that the highly hydrophobic N-terminus of CorS might be involved in temperature-signal perception, the membrane topology of CorS was determined using translational phoA and lacZ fusions, leading to a topological model for CorS with six transmembrane domains (TMDs).

Background: The reduced ability to activate oral tolerance plays a role in the pathogenesis of some gastrointestinal inflammatory diseases. This activation may reflect a preferential reduction of a T-helper (Th)2- or Th3-type response. In recurrent aphthous ulceration (RAU), genetic and environmental factors may contribute to low tolerance, permitting a cytotoxic reaction against the oral epithelium.

[Recognition of internal (TTAGGG)n repeats by telomeric protein TRF1 and its role in maintenance of chromosomal stability in Chinese hamster cells].

Mammalian telomeres contain long tandem (TTAGGG)n repeats, which are protected by a complex of different proteins. Telomeric repeat-binding factors TRF1 and TRF2 play the key role in protection of telomeres through the formation of terminal loops (called T-loop). A T-loop isolates the 3' strand telomeric end and with this mechanism protects telomeres from the influence of enzymes of DNA reparation and telomere fusions and also interferes with the interaction of telomerase with telomeres.

[Conditioning of gray crows (Corvus cornix L.) in a "giant" eight-arm radial maze].

Twelve adult hooded crows (Corvus cornix) were trained to perform a standard radial-maze task in a giant eight-arm outdoor radial maze constructed at the "Chistyi les" Biological Station (Tver oblast) for comparative investigations of spatial memory in birds and mammals. The maze consists of a central part of 250 cm diameter, and has arms of 650 cm length, 170 cm height and 80 cm width. The examined hooded crows showed good task performance.

Protection of internal (TTAGGG)n repeats in Chinese hamster cells by telomeric protein TRF1.

Chinese hamster cells have large interstitial (TTAGGG) bands (ITs) which are unstable and should be protected by an unknown mechanism. Here, we expressed in Chinese hamster V79 cells green fluorescent protein (GFP)-tagged human TRF1, and found that a major fraction of GFP-TRF1 bound to ITs is diffusionally mobile. This fraction strongly decreases after treatment of cells with wortmannin, a protein kinase inhibitor, and this drug also increases the frequency of chromosome aberrations.

Dephosphorylation of histone gamma-H2AX during repair of DNA double-strand breaks in mammalian cells and its inhibition by calyculin A.

The induction of DNA double-strand breaks (DSBs) by ionizing radiation in mammalian chromosomes leads to the phosphorylation of Ser-139 in the replacement histone H2AX, but the molecular mechanism(s) of the elimination of phosphorylated H2AX (called gamma-H2AX) from chromatin in the course of DSB repair remains unknown. We showed earlier that gamma-H2AX cannot be replaced by exchange with free H2AX, suggesting the direct dephosphorylation of H2AX in chromatin by a protein phosphatase. Here we studied the dynamics of dephosphorylation of gamma-H2AX in vivo and found that more than 50% was dephosphorylated in 3 h, but a significant amount of gamma-H2AX could be detected even 6 h after the induction of DSBs.

[Analysis of the effect of a training regime for the correct choice of matched learning in gray crows].

It was shown that a large set of training stimuli promotes abstract concept learning. These experiments were designed to assess whether an application of a large set of training stimuli would facilitate matching learning in crows. Four hooded crows were trained with a set of 72 unique combinations of stimuli in two-alternative simultaneous matching tasks with stimuli of three different categories: achromatic color (white, light-grey, dark-grey, and black), shape (Arabic numerals from 1 to 4 used as visual shapes only), and number of elements (heterogeneous graphic arrays from 1 to 4 items).

A recently described strategy for splice variant specific detection by RT-PCR is based on the use of a primer spanning the junction between exons of the alternative splice form. However, this reaction may generate false-positive results in the presence of excess principal transcript. In this report, transcript variant 3 of T cell immune regulator gene 1 was used as a model to demonstrate a new method to ensure PCR specificity.

A large outdoor radial maze for comparative studies in birds and mammals.

For a comparative neurobiological analysis of spatial learning and memory, a large outdoor eight-arm radial maze was constructed which permits behavioral assessment of many avian and mammalian species both from the laboratory or the wild, using the same metric space and session schedules. It consists of a central part of 250cm diameter, and has arms of 650cm length, 170cm height and 80cm width. In order to determine appropriate training schedules for comparison of different species, we tested four mammalian and two avian species during 9-15 sessions: 18 albino rats (Rattus norvegicus), nine outdoors and nine in a conventional small indoor maze; six guinea pigs (Cavia porcellus); six rabbits (Oryctolagus cuniculus); five hedgehogs (Erinaceus europaeus); seven hooded crows (Corvus corone cornix) and six chickens (Gallus domesticus).

The aim of this study was to further clarify which part of human low density lipoprotein (LDL) is attacked by the MPO/H2O2/Cl- -system and which reactive oxygen species is responsible for the attack. Therefore the influence of this system on the modification of the lipid and protein moiety of LDL was studied in vitro. Using the monochlorodimedone assay it was found that HOCl is produced in micromolar quantities in the absence of LDL and is rapidly consumed by LDL in a concentration dependent manner.

Four gray crows were trained to choose the larger of two sets presented to them within the "1-12" range. The color, form, and size of the stimulus elements on cards, as well as the patterns of their disposition, were varied constantly during training, while at the same time prohibiting the use of extraneous features not relating to the number of elements. In order to exclude choice on the basis of the total area of the elements, series of presentations with variation of the ratio of the number of elements and their total area were carried out: in half of the presentations the larger set consisted of small elements, and the smaller set of large elements; in the second half, the reverse.
