The vector genome (vg) titer measurement, which is used to control patient dosing and ensure control over drug product manufacturing, is essential for the development of recombinant adeno-associated virus (AAV) gene therapy products. While qPCR and droplet digital PCR technologies are commonly implemented for measuring vg titer, chromatographic techniques with UV detectors represent promising future approaches, in line with traditional biotherapeutics. Here, we introduce a novel vg titer measurement approach using size-exclusion high-performance liquid chromatography with UV detection, which achieves excellent method precision (<2% relative SD), demonstrates linearity across a range of concentrations and varied particle content, is stability indicating, and can be bridged with existing vg titer methods.

Background: Methods for matching in longitudinal cohort studies, such as sequential stratification and time-varying propensity scores, facilitate causal inferences in the context of time-dependent treatments that are not randomized where patient eligibility or treatment status changes over time. The tradeoffs in available approaches have not been compared previously, so we compare two methods using simulations based on a retrospective cohort of patients eligible for weight loss surgery, some of whom received it.

Spillover from interparental conflict (IPC) to the parent-child relationship is a risk factor for adolescent emotional, social, and behavioral maladjustment. Parental depression increases the risk for more frequent and intense IPC over periods of months to years, but relatively little is known about whether parental depressive symptoms increase the risk for IPC and/or the propensity for spillover on short timescales. Using daily diary methods, we tested two hypotheses to evaluate whether parental depressive symptoms predict increased risk for IPC spillover: (a) a , in which higher levels of parental depressive symptoms are associated with a greater tendency to experience IPC, which elevates risk for spillover, and (b) a , in which parental depressive symptoms strengthen the within-person linkage between daily couple conflict and poorer parent-adolescent relations.

Parental warmth during the transition from childhood to adolescence is a key protective factor against a host of adolescent problems, including substance use, maladjustment, and diminished well-being. Moreover, adolescents and parents often disagree in their perceptions of parenting quality, and these discrepancies may confer risk for problem outcomes. The current study applies latent profile analysis to a sample of 687 mother-father-6th grade adolescent triads to identify patterns of adolescent-parent convergence and divergence in perceptions of parental warmth.

Recent publicity around the use of new antiobesity medications (AOMs) has focused the attention of patients and healthcare providers on the role of pharmacotherapy in the treatment of obesity. Newer drug treatments have shown greater efficacy and safety compared with older drug treatments, yet access to these drug treatments is limited by providers' discomfort in prescribing, bias, and stigma around obesity, as well as by the lack of insurance coverage.
