p47 -deficient chronic granulomatous disease (p47-CGD) is a primary immunodeficiency caused by mutations in the neutrophil cytosolic factor 1 () gene, resulting in defective NADPH oxidase function in phagocytes. Due to its complex genomic context, the locus is not suited for safe gene editing with current genome editing technologies. Therefore, we developed a targeted coding sequence knock-in by CRISPR-Cas9 ribonucleoprotein and viral vector template delivery, to restore p47 expression under the control of the endogenous locus.

As some proteins are known to interact with sulfated and phosphated biomolecules such as specific glycosaminoglycans, this study derives from the hypothesis that sulfonate and phosphonate groups on solid polymer surfaces might cause specific interfacial interactions. Such surfaces were prepared by plasma polymerization of heptylamine (HA) and subsequent grafting of sulfonate or phosphonate groups via Michael-type addition of vinylic compounds. Adsorption of the proteins fibrinogen, albumin (HSA) and lysozyme on these functionalised plasma polymer surfaces was studied by XPS and quartz crystal microbalance with dissipation (QCM-D).

Antibacterial coating is important to prevent the colonization of medical devices by biofilm forming bacteria that would cause infection and sepsis in patients. Current coating techniques such as immobilization of antimicrobial compounds, time-releasing antibiotic agents and silver nanoparticles, require multiple processing steps, and they have low efficacy and low stability. This study investigated the effectiveness of cure and post-gel shrinkage of three visible light-cured composite resins (In Ten-S [IT], Ivoclar Vivadent; Z100 [ZO], 3M-ESPE; Tetric Ceram [TC], Ivoclar Vivadent) when polymerized with a very high intensity (1296 +/- 2 mW/cm2) halogen light (Astralis 10, Ivoclar Vivadent) for 10 seconds. Irradiation with a conventional (494 +/- 3 mW/cm2) halogen light (Spectrum, Dentsply) for 40 seconds was used for comparison. The effectiveness of cure was assessed by computing the hardness gradient between the top and bottom surfaces of 2-mm composite specimens after curing.
