Article Synopsis
  • Cytogenetic analysis of the product of conception (POC) is crucial for identifying chromosomal abnormalities, which can guide preimplantation genetic testing; traditional G-staining methods often fail due to issues with cell culture.* -
  • Recent research suggests that low-coverage whole-genome sequencing (lcWGS) using next-generation sequencing (NGS) is a promising alternative for POC analysis, with two NGS protocols showing equivalent results across 20 samples.* -
  • However, caution is advised when interpreting data related to the X chromosome, as some anomalies may stem from technical artifacts rather than true genetic conditions, indicating the need for careful analysis in NGS applications.*

The standard treatment for advanced urothelial carcinoma includes platinum-based chemotherapy and programmed cell death protein 1 or programmed death ligand 1 inhibitors. However, urothelial carcinomas are often associated with both intrinsic and acquired resistance to these treatments. Paclitaxel, ifosfamide, and nedaplatin (TIN) chemotherapy has been proven to be effective as the second- or third-line treatment for platinum-resistant advanced urothelial cancer.

Objective: Xq chromosome duplication with complex rearrangements is generally acknowledged to be associated with neurodevelopmental disorders, such as Pelizaeus-Merzbacher disease (PMD) and MECP2 duplication syndrome. For couples who required a PGT-M (pre-implantation genetic testing for monogenic disease) for these disorders, junction-specific PCR is useful to directly detect pathogenic variants. Therefore, pre-clinical workup for PGT-M requires the identification of the junction of duplicated segments in PMD and MECP2 duplication syndrome, which is generally difficult.

Article Synopsis
  • GATA4 is linked to congenital heart disease and now recognized for its role in disorders of sexual development (DSD), particularly in a case of a 46,XY DSD patient with an atrial septal defect.
  • A specific mutation (c.487C>T) was found to reduce transcriptional activity of downstream genes, showing that heart development may need stricter GATA4 regulation than testicular development.
  • The study suggests that GATA4 variants exhibit a dominant-negative effect, impacting transcriptional activity when mixed with normal protein, and indicates that some pathogenic variants could also be found in healthy parents, hinting at a complex genetic threshold theory for these conditions.

Background: In all the prefectures of Japan, with the exception of Shiga Prefecture, more than half of local governments have an organized prostate-specific antigen (PSA) screening system in place. However, in the Shiga Prefecture, only a single city performed PSA screening over the time period of this survey. The purpose of the present study was to determine the clinical, pathological, and therapeutic features of newly diagnosed prostate cancer in localities where a formally organized screening system was almost entirely absent.

We evaluated the safety of laparoscopic radical cystectomy (LRC) during initial phases and its learning curve in a Japanese multicenter cohort by studying 436 patients who underwent LRC with no robot assistance at 10 institutions in Japan. We divided the patients into three groups according to cumulative surgical volume at each institution (first 10 cases, 11-30 cases, after 31 cases in each institution), and compared perioperative and pathologic variables among the three groups. The first, second, and third groups included 100, 166, 170 patients, respectively.

Article Synopsis
  • The study analyzed the outcomes of laparoscopic adrenalectomy in 67 patients with adrenal metastasis across 11 hospitals from 2003 to 2017, focusing on survival benefits and complications.
  • The most common primary cancers were lung cancer and renal cell carcinoma, with a median overall survival of 3.8 years; intraoperative and postoperative complication rates were both around 10%.
  • Key findings indicated that larger tumor size, presence of extra-adrenal metastasis before and at the time of surgery, and positive surgical margins were associated with shorter overall survival, suggesting that careful patient selection is crucial for better surgical outcomes.

Guard cells are indispensable for higher plants because they control gas exchange and water balance to maintain photosynthetic activity. The signaling processes that govern their movement are controlled by several factors, such as abscisic acid (ABA), blue light, pathogen-associated molecular patterns (PAMPs), and carbon dioxide. Herein, we demonstrated that the amino acid glutamate (Glu), a well-known mammalian neurotransmitter, functions as a novel signaling molecule in stomatal closure in both Arabidopsis and fava bean (Vicia faba L.

Nitric oxide (NO) is a ubiquitous signaling molecule involved in diverse physiological processes, including plant senescence and stomatal closure. The NO and cyclic GMP (cGMP) cascade is the main NO signaling pathway in animals, but whether this pathway operates in plant cells, and the mechanisms of its action, remain unclear. Here, we assessed the possibility that the nitrated cGMP derivative 8-nitro-cGMP functions in guard cell signaling.
