Article Synopsis
  • This study developed advanced methods (UPLC-MS/MS, HPLC, and anthrone colorimetry) to analyze the chemical components in two plant species, Puerariae Thomsonii Radix and Puerariae Lobatae Radix.
  • The research found 17 flavonoids in both plants and highlighted significant differences in their chemical compositions, specifically showing lower cellulose and higher starch in Puerariae Thomsonii compared to Puerariae Lobatae.
  • The methods established are noted for being simple, rapid, accurate, and sensitive, providing a foundation for future utilization and evaluation of these medicinal plants.

Puerarin was conjugated with bovine serum albumin(BSA) and ovalbumin(OVA) by periodate oxidation to serve as the immunogen and coating antigen, respectively. BALB/c mice were immunized with puerarin-BSA according to the routine immunization procedure, and the titer and specificity of serum were detected after three immunization. After booster immunization, mouse spleen lymphocytes were fused with mouse myeloma cells, and 24 hybridoma cell lines of the monoclonal antibodies against puerarin were screened by monoclonal antibody screening technique.
