The cells of hematopoietic and vascular endothelial cell lineages are believed to share a common precursor, termed hemangioblast. However, the existence of a growth factor acting relatively specifically on hemangioblasts remains unclear. Here we report the identification of hemangiopoietin (HAPO), a novel growth factor acting on both hematopoietic and endothelial cell lineages.

An MRI method for quantification of cerebral blood volume (CBV) in time-course studies of angiogenesis is described. Angiogenesis was stimulated by acclimation to hypoxia. The change in relaxation rate, R2, which is relatively sensitive to the microvasculature, was quantified before and after infusion of a superparamagnetic vascular contrast agent (MION).

Dual X-ray absorptiometry (DXA) has currently become a clinical standard for the assessment of bone mass and bone mineral density (BMD) at multiple sites for the diagnosis and follow-up assessment of osteoporosis in humans. The precision of DXA measurement in human studies has been well documented during the last two decades. However, there have been no systematic reports on the precision and accuracy of BMD measurements in mice using DXA, although mice have proven to be useful models for the study of osteoporosis.

Previous data have demonstrated that CXC-chemokine platelet factor 4 (PF4) inhibits the proliferation of the human erythroleukemia cell line (HEL). However, the mechanism of action is unclear at present. The signaling transduction induced by PF4 in the HEL was compared with that induced by transforming growth factor beta1 (TGF-beta1), which is also a potent inhibitor of HEL growth.

Article Synopsis
  • PTH treatment led to a significant increase in bone mineral density (BMD) in C57BL/6J mice, particularly in the tibia and femur, with observable effects within 1-2 weeks and continuing increases over 7 weeks.
  • The study also showed that while both trabecular and cortical bone parameters improved with PTH treatment, the effects were less pronounced in the lumbar vertebrae, where significant changes only appeared after 7 weeks.
  • Increased gene expression related to bone formation and turnover was noted in the long bones, indicating an anabolic action of PTH at both the tissue and cellular levels.
