While poly (adenosine diphosphate-ribose) polymerase inhibitors (PARPi) have achieved specific clinical benefits in a subset of pancreatic ductal adenocarcinoma (PDAC) patients, the potential role of the PARPi niraparib in PDAC necessitates further exploration. In this study, we demonstrated that Niraparib exhibited a pronounced inhibitory effect on autophagy in PDAC both in vitro and in vivo. Mechanistically, this inhibition was primarily attributed to niraparib's ability to disrupt the fusion process between autophagosomes and lysosomes, while potentially exerting a relatively minor impact on the initial stage of autophagy.

Despite the advances in antibody-guided cell typing and mass spectrometry-based proteomics, their integration is hindered by challenges for processing rare cells in the heterogeneous tissue context. Here, we introduce Spatial and Cell-type Proteomics (SCPro), which combines multiplexed imaging and flow cytometry with ion exchange-based protein aggregation capture technology to characterize spatial proteome heterogeneity with single-cell resolution. The SCPro is employed to explore the pancreatic tumor microenvironment and reveals the spatial alternations of over 5000 proteins by automatically dissecting up to 100 single cells guided by multi-color imaging of centimeter-scale formalin-fixed, paraffin-embedded tissue slide.

In this paper, we focus on eddy current array (ECA) technology for defect detection in finely grooved structures of spinning cylinders, which are significantly affected by surface texture interference, lift-off distance, and mechanical dither. Unlike a single eddy current coil, an ECA, which arranges multiple eddy current coils in a specific configuration, offers not only higher accuracy and efficiency for defect detection but also the inherent properties of space and time for signal acquisition. To efficiently detect defects in finely grooved structures, we introduce a spatiotemporal self-attention mechanism to ECA testing, enabling the detection of defects of various sizes.
