Background And Purpose: Anti-agrin antibodies (agrin Abs) have recently been identified in patients with myasthenia gravis (MG), sometimes in conjunction with antibodies (Abs) to the acetylcholine receptor (AChR), muscle-specific tyrosine kinase (MuSK), or low-density lipoprotein receptor-related protein 4. This study aimed to develop an in-house cell-based assay (CBA) for detecting agrin Abs, and to test its application to serum samples collected from individuals diagnosed with MG.

Methods: Agrin complementary DNA as cloned into a pCMV6-AC-GFP vector, which was subsequently transfected into human embryonic kidney 293T (HEK293T) cells.

Based on the potent and selective antagonism exhibited by truncated North (N)-methanocarba adenosine analogs, we synthesized a series of 5'-deoxy (N)-methanocarba nucleosides to explore their structure-activity relationships (SAR). The stereoselective synthesis of the North cyclopropyl-fused alcohol was achieved from d-ribose using ring-closing metathesis, oxidative rearrangement, and cyclopropanation as key steps. Mitsunobu reactions were employed to condense nucleobases with glycosyl donors, followed by N functionalization with various amines.

Sorghum () is a gluten-free supercrop with a high content of phenolic compounds, along with anti-nutrient factors such as tannin that limit its use in food. In this study, we conducted solid-state fermentation for sorghum with to reduce the tannin content and value-added sorghum by enhancing biological properties. The results showed that fermented sorghum had 1.

Background: Preservative-free chloroprocaine is a promising spinal anesthetic for ambulatory surgeries, offering a short duration of action and minimal side effects, which promote faster recovery and discharge. Thus, this study aimed to compare chloroprocaine hydrochloride to the widely used bupivacaine as a spinal anesthetic in ambulatory anorectal surgeries. We hypothesized that chloroprocaine will lead to quicker recovery and discharge, supporting its use in the ambulatory surgical setting.

We hypothesized a treatment guideline for ulna/radius nonunion after failed surgical treatment and propose to verify it in a prospective study. Herein, we report our preliminary findings and review the current trend.  Six patients who met the criteria were retrospectively investigated, and we further categorized "nonunion of both the radius/ulna" into four subcategories.

Article Synopsis
  • - The immune system relies on both innate and adaptive responses to fight pathogens, and individuals with weakened immune systems require effective methods to enhance their immune function.
  • - This study investigates the effects of the probiotic OTG1204 on macrophages, crucial for immune defense, specifically examining its potential to stimulate the immune response in conditions of immunosuppression induced by cyclophosphamide (CTX).
  • - Results showed that OTG1204 activated macrophages, increasing important signaling molecules and cytokines through specific pathways, and helped restore immune function in immunosuppressed mice by improving immune cell populations and overall health indicators.

This study investigated the impact of conformation on the binding affinity of carbanucleosides to A and A adenosine receptors (ARs). A series of nucleosides, including saturated, unsaturated, North (N)-methano, and South (S)-methanocarbanucleosides was prepared, and their binding affinities to AAR and AAR were assessed. Biological evaluations revealed that all synthesized (S)-methanocarbanucleosides had negligible binding to both receptors, and most (N)-methanocarbanucleosides exhibited high binding affinities.

Background And Purpose: Public medical insurance data are increasingly used to study myasthenia gravis (MG); however, a validated case definition is lacking. We assessed the clinical characteristics of patients identified according to previously used case definitions.

Methods: Patients with diagnosis code G70.

Article Synopsis
  • Researchers studied the quality of life (QOL) of patients with a condition called MOGAD and compared it to another condition called AQP4-IgG positive NMOSD.
  • They found that the overall QOL was similar between both groups, but many patients in both groups struggled with sleep, pain, and feelings of sadness.
  • The study suggests that both conditions need more focus on improving sleep and mental health to help patients feel better overall.

Objective: We analyze the relationship between employment status, income, and sleep in South Africa to address two research questions: (1) How does employment status influence the sleep quantity of the individual and their partner? (2) How does income impact the sleep quantity of the individual and their partner?

Method: Using data from 1600 Black African couples in the South African Time Use Study, we employ the Actor-Partner Interdependence Model to investigate the relationship between employment status, income, and sleep in couples. We categorize nighttime sleep into three categories: recommended sleep (7-9 hours), short sleep (<7 hours), and long sleep (>9 hours).

Results: Employed individuals were more likely to get the recommended amount of sleep and less likely to experience long sleep.

  • - The study focuses on spinal muscular atrophy (SMA), a genetic disease that results in the loss of motor neurons, leading to muscle weakness, and examines the diagnostic process for Korean SMA patients over 23 years.
  • - In a review of 38 patients, it was found that the average age for symptom onset was 3 years, but diagnosis occurred much later at 25 years, with a notable delay of approximately 19.6 years, particularly longer for type III SMA patients.
  • - The research emphasizes the importance of improving diagnostic accuracy for SMA as treatment options evolve, marking a significant exploration of the diagnostic journey for patients in Korea.

The gut microbial and metabolic characteristics of intestinal Behçet's disease (BD), a condition sharing many clinical similarities with ulcerative colitis (UC) and Crohn's disease (CD), are largely unexplored. This study investigated the gut microbial and metabolic characteristics of intestinal BD as well as potential biomarkers, comparing them with those in UC, CD, and healthy controls. Colon tissue and stool samples from 100 patients (35 UC, 30 CD, and 35 intestinal BD) and 41 healthy volunteers were analyzed using 16S ribosomal RNA sequencing to assess microbial diversity, taxonomic composition, and functional profiling.

  • The study explored the link between narrowing of cerebral arteries (stenosis) and depressive symptoms in elderly patients over 65 years old, using data from a psychiatric clinic over nearly two decades.
  • Among 365 participants, 29.6% had cerebral artery stenosis, and those with stenosis had higher depression scores on the Geriatric Depression Scale (GDS-15).
  • Specifically, bilateral middle cerebral artery (MCA) stenosis and left posterior cerebral artery (PCA) stenosis were significantly associated with greater depressive symptoms, with bilateral MCA stenosis indicating a stronger correlation with depression severity.

The tumor microenvironment comprises both tumor and non-tumor stromal cells, including tumor-associated macrophages (TAMs), endothelial cells, and carcinoma-associated fibroblasts. TAMs, major components of non-tumor stromal cells, play a crucial role in creating an immunosuppressive environment by releasing cytokines, chemokines, growth factors, and immune checkpoint proteins that inhibit T cell activity. During tumors develop, cancer cells release various mediators, including chemokines and metabolites, that recruit monocytes to infiltrate tumor tissues and subsequently induce an M2-like phenotype and tumor-promoting properties.

B cell receptors (BCRs) denote antigen specificity, while corresponding cell subsets indicate B cell functionality. Since each B cell uniquely encodes this combination, physical isolation and subsequent processing of individual B cells become indispensable to identify both attributes. However, this approach accompanies high costs and inevitable information loss, hindering high-throughput investigation of B cell populations.
