Paediatric medulloblastomas with chromothripsis are characterised by high genomic instability and are among the tumours with the worst prognosis. However, the molecular makeup and the determinants of the aggressiveness of chromothriptic medulloblastoma are not well understood. Here, we apply spatial transcriptomics to profile a cohort of 13 chromothriptic and non-chromothriptic medulloblastomas from the same molecular subgroup.

Article Synopsis
  • The study aimed to determine if eating during labor affects outcomes by comparing food consumption to clear fluids in laboring patients.
  • 129 healthy patients were randomized into two groups: one allowed to eat and the other restricted to clear fluids, with various maternal and neonatal outcomes measured.
  • Results showed no significant differences in complications, labor progression, or outcomes between the two groups, leading to the conclusion that eating during labor is generally safe but should be approached cautiously based on individual patient risk factors.

Low-abundance members of microbial communities are difficult to study in their native habitats, including Escherichia coli, a minor but common inhabitant of the gastrointestinal tract, and key opportunistic pathogen of the urinary tract. While multi-omic analyses have detailed interactions between uropathogenic Escherichia coli (UPEC) and the bladder mediating urinary tract infection (UTI), little is known about UPEC in its pre-infection reservoir, the gastrointestinal tract, partly due to its low relative abundance (<1%). To sensitively explore the genomes and transcriptomes of diverse gut E.

Article Synopsis
  • Cancer-specific T cells often fail to eliminate tumors, so researchers explored using CD4 T cell receptors (CD4TCRs) from tumor-bearers for new treatments.
  • They focused on TCRs that target unique neoantigens presented by the surrounding tissue of MHC class II-negative cancer cells, testing the 11 most common TCRs.
  • Some TCRs shared similar structures despite different sequences, effectively attacking tumors and reversing tumor progression; choosing TCRs based on their genetic similarities may enhance therapeutic success.

Purpose: Fetal sex is independently associated with pregnancy complications and impacts neonatal outcomes. Evidence suggests that females have an advantage over males, with a better outcome in the perinatal period. In addition, fetal outcome in twin gestations is also related to the intrauterine position of the fetus, such as the first, the presenting or second twin.

Low-abundance members of microbial communities are difficult to study in their native habitats. This includes , a minor, but common inhabitant of the gastrointestinal tract and opportunistic pathogen, including of the urinary tract, where it is the primary pathogen. While multi-omic analyses have detailed critical interactions between uropathogenic (UPEC) and the bladder that mediate UTI outcome, comparatively little is known about UPEC in its pre-infection reservoir, partly due to its low abundance there (<1% relative abundance).

Aberrant TGFβ signaling is linked to metastasis and tumor immune escape of many cancers including metastatic triple negative breast cancer (mTNBC). Previously, we have found that oncolytic adenoviruses expressing a TGFβ signaling inhibitory protein (sTGFβRIIFc) induced immune activation in a mouse TNBC (4T1) immunocompetent subcutaneous model with intratumoral injection. Systemic administration of adenoviruses can be a superior route to treat mTNBC but faces the challenges of increased toxicity and viral clearance.

Article Synopsis
  • Women with endometriosis experience altered pain perceptions, leading to different pain management needs after cesarean delivery (CD).
  • A study compared post-operative pain scores and analgesic use between women with and without endometriosis, finding that those with endometriosis required higher doses of pain medication on the first post-operative day.
  • Despite increased analgesic use in the endometriosis group, their pain intensity, measured on a visual analogue scale, did not differ significantly from the control group.

Purpose: To achieve eradication of solid tumors, we examined how many neoantigens need to be targeted with how many T-cell receptors (TCR) by which type of T cells.

Experimental Design: Unmanipulated, naturally expressed (autochthonous) neoantigens were targeted with adoptively transferred TCR-engineered autologous T cells (TCR-therapy). TCR-therapy used CD8+ T-cell subsets engineered with TCRs isolated from CD8+ T cells (CD8+TCR-therapy), CD4+ T-cell subsets engineered with TCRs isolated from CD4+ T cells (CD4+TCR-therapy), or combinations of both.
