Intermetallics, which encompass a wide range of compounds, often exhibit similar or closely related crystal structures, resulting in various intermetallic systems with structurally derivative phases. This study examines the hypothesis that deformation behavior can be transferred from fundamental building blocks to structurally related phases using the binary samarium-cobalt system. SmCo and SmCo are investigated as fundamental building blocks and compared them to the structurally related SmCo and SmCo phases.

Glioblastoma cells rely on connexin 43 (Cx43)-based gap junctions (GJs) for intercellular communication, enabling them to integrate into a widely branched malignant network. Although there are promising prospects for new targeted therapies, the lack of clinically feasible GJ inhibitors has impeded their adoption in clinical practice. In the present study, we investigated tonabersat (TO), a blood-brain-barrier-penetrating drug with GJ-inhibitory properties, in regard to its potential to disassemble intercellular connectivity in glioblastoma networks.

Mammalian ribosomal RNA (rRNA) molecules are highly abundant RNAs, decorated with over 220 rRNA modifications. Previous works have shown that some rRNA modification types can be dynamically regulated; however, how and when the mammalian rRNA modification landscape is remodeled remains largely unexplored. Here, we employ direct RNA sequencing to chart the human and mouse rRNA epitranscriptome across tissues, developmental stages, cell types, and disease.

We describe an algorithm for generating fiber-filled volume elements for use in computational homogenization schemes which accounts for a coupling of the fiber-length and the fiber-orientation. For prescribed fiber-length distribution and fiber-orientation tensor of second order, a maximum-entropy estimate is used to produce a fiber-length-orientation distribution which mimics real injection molded specimens, where longer fibers show a stronger alignment than shorter fibers. We derive the length-orientation closure from scratch, discuss its integration into the sequential addition and migration algorithm for generating fiber-filled microstructures for industrial volume fractions and investigate the resulting effective elastic properties.

Background: While several studies have explored the outcomes of transcatheter interventions for modified Blalock-Taussig shunts (MBTSs) in a broad range of congenital heart diseases, none have specifically examined the interventions in patients with hypoplastic left heart syndrome (HLHS) who underwent Norwood palliation (NP).

Methods: This retrospective study was conducted between 2020 and 2024, when 24 urgent interventions were performed on 17 patients at our center. We recorded several key outcomes, including early and late intervention-related complications, the need for reintervention, the interval between the NP and the first intervention, shunt patency following the intervention, associated morbidities, and thrombosis-related sudden events.

  • Access to abortion care in Germany is hindered by various barriers, such as geographical issues, confidentiality concerns, waiting times, stigmatization, lack of information, and costs.
  • A survey of 594 women who terminated unwanted pregnancies revealed that 80.1% experienced at least one barrier, with significant percentages reporting multiple barriers related to social stigma and privacy concerns.
  • To improve access to safe abortions, the study suggests implementing decriminalization, enhancing care structures, providing financial support, and reducing stigma and discrimination.

Background And Purpose: Despite recent advances of online image-guided high-precision patient positioning and adaptation using magnetic resonance imaging (MRI) or cone-beam computed tomography (CT), standard radiation therapy pathway still involves a dedicated simulation scan. The aim of this study was to evaluate the feasibility and planning quality of integrating a simulation-free treatment planning workflow for adaptive online MRI-guided radiation therapy on a 1.5 T MRI linear accelerator (MRI-Linac) in prostate cancer using diagnostic CT (dCT) scans.

Background: Invasive vagus nerve stimulation (iVNS) is approved for the treatment of major depressive disorder (MDD). The limited understanding of its underlying mechanisms, however, hinders stratification and the prediction of treatment response. Given the strong projections of the afferent vagal nucleus to brain regions involved in emotional processing, we tested whether acute transauricular VNS (taVNS) can improve emotional processing that is a core deficit in MDD.

Objective: Although the operating theatre offers unique didactic opportunities, it can be perceived as an uncomfortable environment by medical students due to the lack of theatre etiquette, time pressure and parallel work of different disciplines. We investigated whether virtual reality (VR) training can significantly reduce some of these fears and improve surgical education for medical students.

Methods: We randomly divided a group of 24 medical students and investigated the effects of a VR application (digitally recreated operating theatre tour) and compared it with currently-in-use written instructions for operational tasks in the operating theatre.

Background: Metabolic bone disease (MBD) of newborns (NB) is characterized by tissue reduction and inadequate postnatal bone mineralization with clinical, laboratory, and/or radiologic repercussions between the third and twelfth weeks of postnatal life, which, in its most severe forms, can lead to a growth deficit and fractures. The aim of our study is to evaluate the incidence of MBD in premature patients receiving parenteral nutrition for >15 days in the neonatal intensive care unit (NICU) and assess their clinical and laboratory characteristics.

Methods: Single-center retrospective cohort study.

Article Synopsis
  • * A retrospective analysis of 19 patients showed significant improvement in blood oxygen saturation after PVB, with many experiencing positive changes in pulmonary valve and artery measurements, although some faced recurrent desaturation requiring further interventions.
  • * The findings suggest that PVB is a safe and effective early palliative measure for managing cyanosis in TOF patients, allowing for improved pulmonary development and delaying the need for more invasive surgical repairs, with BRCTs not significantly altering surgical approaches.

Purpose: Prophylactic insertion of an external ventricular drainage (EVD) prior to the resection of posterior fossa metastases (PFMs) is a common approach to address postoperative transient and permanent hydrocephalus. However, predicting surgery-related hydrocephalus in the preoperative phase continues to be a challenge. This study aims to analyze the incidence, preoperatively collectable risk factors and necessity of perioperative external ventricular drainage placement after posterior fossa metastasis surgery.

The development of macrocyclic binders to therapeutic proteins typically relies on large-scale screening methods that are resource-intensive and provide little control over binding mode. Despite considerable progress in physics-based methods for peptide design and deep-learning methods for protein design, there are currently no robust approaches for design of protein-binding macrocycles. Here, we introduce RFpeptides, a denoising diffusion-based pipeline for designing macrocyclic peptide binders against protein targets of interest.

The complex kinetics of disease-related amyloid aggregation of proteins such as α-Synuclein (α-Syn) in Parkinson's disease and Aβ42 in Alzheimer's disease include primary nucleation, amyloid fibril elongation and secondary nucleation. The latter can be a key accelerator of the aggregation process. It has been demonstrated that the chaperone domain BRICHOS can interfere with the secondary nucleation process of Aβ42.

Article Synopsis
  • Liver fibrosis results from chronic liver inflammation leading to excess extracellular-matrix production by activated hepatic stellate cells (HSCs), which can lead to serious liver damage.* -
  • Research on mice showed that lacking HIF-prolyl-hydroxylase 1 (PHD1) reduced liver fibrosis severity and related inflammation, while treatment with the inhibitor DMOG did not prevent fibrosis.* -
  • PHD1 deficiency was linked to decreased activation of HSCs and lower expression of fibrotic and inflammatory markers, suggesting that targeted PHD1 inhibitors might be effective in managing liver fibrosis.*

Central nervous system diseases negatively affect patients and society. Providing successful noninvasive treatments for these diseases is challenging because of the presence of the blood-brain barrier. While protecting the brain's homeostasis, the barrier limits the passage of almost all large-molecule drugs and most small-molecule drugs.

Article Synopsis
  • Conventional image-guided radiotherapy (conv-IGRT) for prostate cancer fails to account for anatomical changes between treatment sessions, while online-adaptive magnetic resonance guided radiotherapy (OA-MRgRT) has potential for better sparing of critical organs and improved targeting.
  • The study analyzed dose accumulation in ten prostate cancer patients treated with OA-MRgRT versus conv-IGRT, using advanced imaging techniques for dose mapping.
  • Results showed that while both methods yielded similar target dose parameters, OA-MRgRT delivered lower doses to organs-at-risk like the rectum and bladder compared to conv-IGRT, though these differences didn't lead to clinically significant outcomes.

Polypoidal choroidal vasculopathy (PCV) is an aneurismal type of macular neovascularization that show similarities with age-related macular degeneration and diseases that are part of the pachychoroid disease spectrum. Exudative changes in PCV can be treated with intravitreal anti-vascular endothelial growth factor monotherapy; however, a combination therapy with photodynamic therapy may be required. In this systematic review and meta-analysis, we evaluated the efficacy of faricimab for PCV.

Background: People with cystic fibrosis carrying two nonsense alleles lack CFTR-specific treatment. Growing evidence supports the hypothesis that nonsense mutation identity affects therapeutic response, calling for mutation-specific CF models. We describe a novel W1282X mouse model and compare it to an existing G542X mouse.
