Although the epidermal growth factor receptor 2 (ErbB2) and Notch1 signaling pathways have both significant roles in regulating cardiac biology, their interplay in the heart remains poorly investigated. Here, we present evidence of a crosstalk between ErbB2 and Notch1 in cardiac cells, with effects on autophagy and proliferation. Overexpression of ErbB2 in H9c2 cardiomyoblasts induced Notch1 activation in a post-transcriptional, p38-dependent manner, while ErbB2 inhibition with the specific inhibitor, lapatinib, reduced Notch1 activation.

JCO We present the final analysis of the phase III noninferiority, randomized ShortHER trial comparing 9 weeks versus 1 year of adjuvant trastuzumab with chemotherapy in patients with human epidermal growth factor receptor 2-positive (HER2+) early breast cancer (BC). Women with HER2+ BC were randomly assigned to anthracycline-taxane combinations plus 1-year trastuzumab (arm A, long) or 9-week trastuzumab (arm B, short). Here, we report the second coprimary end point overall survival (OS), updated disease-free survival (DFS), and outcomes according to hormone receptor status, age, and nodal status.

Pathological complete response (pCR) after neoadjuvant chemotherapy (NAC) is a strong prognostic factor in breast cancer (BC). The aim of this study was to investigate whether semiquantitative parameters derived from baseline [F]Fluorodeoxyglucose ([F]FDG) positron emission computed tomography/computed tomography (PET/CT) could predict pCR after NAC and survival outcomes in patients affected by different molecular subtypes of BC. In this study, site-specific natural background levels (NBLs) were determined for 18 elements (Al, As, Be, Cd, Co, Cu, Cr, Fe, Hg, K, Mn, Mo, Ni, Pb, Tl, U, V, and Zn) in two sediment cores collected offshore the Bagnoli-Coroglio brownfield site (Gulf of Pozzuoli, southern Italy) to accurately assess the degree of contamination and the historical trends in Heavy Metals (HMs) enrichment. This objective was pursued taking in account the high temporal and spatial variability of the geochemical properties of the area due to the local geothermal activity. Coastal lagoons are important but sensitive environments, being transitional zones between land and sea. The Khnifiss lagoon is the most important desert wetland in Morocco, but little data have been produced concerning heavy metal geochemistry and enrichments in the sediments. Baseline study of natural (Be, Pb, Ra, Th, Ra, K) and anthropogenic (Cs) radionuclides was carried out in two cores collected from Sabaki River estuary (Kenya, Indian Ocean). There was no exponential decrease of excess Pb down the cores, which did not allow dating and determination of heavy metal pollution history. The use of Cs as a time marker was not possible due to its low fallout rates in East Africa.

Background: The combination of a microtubule inhibitor (eribulin) with a nucleoside analog (gemcitabine) may synergistically induce tumor cell death, particularly in triple negative breast cancer (TNBC) characterized by high cell proliferation, aggressive behavior, and chemo-resistance.

Isotopic composition of mercury (Hg) in marine organisms and sediment cores was used to identify sources and reconstruct historical trends of contamination in the coastal-marine area of Rosignano Solvay (Italy), affected by Hg pollution from a chlor-alkali plant on the near land. Sediments show a wide range of Hg concentration and Hg isotope signatures. Particularly, coupled Hg concentration and δHg values trace inputs from different sources.

Laboratories from 14 countries (with different levels of expertise in radionuclide measurements and Pb dating) participated in an interlaboratory comparison exercise (ILC) related to the application of Pb sediment dating technique within the framework of the IAEA Coordinated Research Project. The laboratories were provided with samples from a composite sediment core and were required to provide massic activities of several radionuclides and an age versus depth model from the obtained results, using the most suitable Pb dating model. Massic concentrations of Zn and Cu were also determined to be used for chronology validation.

Background: Different studies suggest that fulvestrant 500 mg every 28 days (HD-FUL) could be an active treatment in HR+ advanced breast cancer (ABC) patients even treated with aromatase inhibitors in the adjuvant setting. Background: NEPA is an oral fixed-dose combination of netupitant, a new highly selective neurokinin-1 receptor antagonist, and palonosetron. This study was conducted to evaluate whether the efficacy of NEPA against chemotherapy-induced nausea and vomiting (CINV) in cycle 1 would be maintained over subsequent chemotherapy cycles in breast cancer patients receiving adjuvant anthracycline plus cyclophosphamide (AC). The study also describes the relationship between efficacy on day 1 through 5 (overall period) and control of CINV on day 6 through 21 (very late period) in each cycle.

Pb and Cs vertical profiles in coastal sediments are studied using a sedimentation-mixing model. The cores were sampled in a complex coastal area characterized by the presence of different impact sources and environmental uses (riverine inputs, commercial and military harbour, marine protected area) in the Eastern Ligurian Sea, Italy. The analysis of accumulation and dispersion processes is performed using a numerical advection and diffusion model, in terms of the independent variables - time and mass depth.

The compartment model POSEIDON-R with an embedded dynamic food web model was used to assess Cs distributions in the Mediterranean and Black Seas during 1945-2020 due to the weapon testing and accident at the Chernobyl nuclear power plant. Three maximums of contamination of surface waters can be identified from 1950 in the Mediterranean Sea system. Two of them (in 1959 and 1963) were caused by atmospheric deposition due to the nuclear weapon testing.

Lessons Learned: The androgen receptor (AR) is present in most breast cancers (BC), but its exploitation as a therapeutic target has been limited.This study explored the activity of dehydroepiandrosterone (DHEA), a precursor being transformed into androgens within BC cells, in combination with an aromatase inhibitor (to block DHEA conversion into estrogens), in a two-stage phase II study in patients with AR-positive/estrogen receptor-positive/human epidermal growth receptor 2-negative metastatic BC.Although well tolerated, only 1 of 12 patients obtained a prolonged clinical benefit, and the study was closed after its first stage for poor activity.

In continental margins, canyons appear to act as natural conduits of sediments and organic matter from the shelf to deep basins, providing an efficient physical pathway for transport and accumulation of particles with their associated land-produced contaminants. However, these mechanisms have not been yet sufficiently explored by geochemical markers. The continental slope of the south Sardinia has been used as a natural laboratory for investigating mechanisms and times of transfer dynamics of contaminants from land to sea and from shelf to deep sea through an articulated system of submarine canyons.
