Article Synopsis
  • Regulation of dopamine activity is crucial for treating conditions like schizophrenia, with LB-102 being a new dopamine D/5-HT inhibitor aimed at this purpose.
  • The study measured how effectively LB-102 occupied dopamine receptors in healthy volunteers using PET scans at various doses, finding that 50 mg consistently provided the desired occupancy over 24 hours.
  • Results indicated that while steady dosing led to consistent receptor occupancy, the relationship between receptor occupancy and plasma concentrations was different than usual, and LB-102 was safe and well-tolerated across all tested doses.

Background: It is not known whether the transition from obesity and severe obesity, as 2 different metabolic disease entities, affect flow-mediated and, thus, endothelium-dependent epicardial vasodilation.

Objectives: The purpose of this study was to investigate the effect of obesity and severe obesity on flow-mediated epicardial vasomotion with positron emission tomography/computed tomography-determined longitudinal decrease in myocardial blood flow (MBF) from the base-to-apex direction of the left ventricle or gradient.

Methods: N-ammonia positron emission tomography/computed tomography evaluated global MBF during pharmacologically induced hyperemia and at rest for assessment of coronary microvascular function.

Purpose Of The Review: Cardiac involvement in systemic sarcoidosis or isolated cardiac sarcoidosis plays a pivotal role in the clinical manifestation and prognostication. Active-inflammatory cardiac sarcoidosis is associated with a regional impairment of coronary microvascular function that may confer further detrimental effects on myocardial function needing further characterization.

Recent Findings: Clinical investigations with cardiac positron emission tomography/computed tomography in conjunction with F-fluorodeoxyglucose to determine myocardial inflammation and N-ammonia to quantify myocardial blood flow (MBF) in patients with known or suspected cardiac sarcoidosis outlined that sarcoidosis-induced myocardial inflammation was associated with adverse effects on corresponding regional coronary microvascular function.

Similar to soils, tree stems emit and consume nitrous oxide (NO) from the atmosphere. Although tree leaves dominate tree surface area, they have been completely excluded from field NO flux measurements and therefore their role in forest NO exchange remains unknown. We explored the contribution of leaf fluxes to forest NO exchange.

Greenhouse gas (GHG) fluxes from peatland soils are relatively well studied, whereas tree stem fluxes have received far less attention. Simultaneous year-long measurements of soil and tree stem GHG fluxes in northern peatland forests are scarce, as previous studies have primarily focused on the growing season. We determined the seasonal dynamics of tree stem and soil CH, NO and CO fluxes in a hemiboreal drained peatland forest.

Positron emission tomography (PET) myocardial perfusion imaging in conjunction with tracer-kinetic modeling enables the concurrent assessment of myocardial perfusion and regional myocardial blood flow (MBF) of the left ventricle in absolute terms in milliliters per gram per minute (mL/g/min). The non-invasive quantification of MBF during pharmacologically induced hyperemia, at rest, and corresponding myocardial flow reserve (MFR) opens a new avenue for the identification and characterization of classical or endogen type of coronary microvascular dysfunction (CMD) as functional substrate for microvascular angina in patients with non-obstructive coronary artery disease (CAD) and/or no CAD at all. Further, PET-MBF quantification expands the scope of conventional myocardial perfusion imaging from the identification of advanced, and flow-limiting, epicardial CAD to early stages of atherosclerosis and/or CMD.

Article Synopsis
  • - Digital PET/CT systems with a long axial field of view are the latest advancements, offering wider anatomical coverage and enhanced system sensitivity for better imaging.
  • - Early findings indicate improvements in image quality and quantification, while also allowing for low-dose exams and shorter acquisition times, particularly in studying cardiac physiology.
  • - This technology enables comprehensive assessment of multiple organs and vascular structures, presenting new possibilities for understanding systemic disorders and their effects on cardiovascular health.

Article Synopsis
  • The study aims to evaluate the effectiveness of O-water PET myocardial perfusion imaging in diagnosing coronary artery disease (CAD) by comparing it to established methods like invasive coronary angiography (ICA) and coronary computed tomography angiogram (CCTA).
  • O-water is noted for its high accuracy in measuring blood flow in the heart, but its clinical use has been limited due to complex production and analysis requirements.
  • The RAPID WATER FLOW study will involve around 215 participants from multiple sites and will focus on O-water PET’s sensitivity and specificity in detecting significant blockages, also considering specific populations like women, obese, and diabetic individuals.

Histosols cover about 8-10% of Lithuania's territory and most of this area is covered with nutrient-rich organic soils (Terric Histosols). Greenhouse gas (GHG) emissions from drained Histosols contribute more than 25% of emissions from the Land Use, Land Use Change and Forestry (LULUCF) sector. In this study, as the first step of examining the carbon dioxide (CO) fluxes in these soils, total soil CO efflux and several environmental parameters (temperature of air and topsoil, soil chemical composition, soil moisture, and water table level) were measured in drained Terric Histosols under three native forest stands and perennial grasslands in the growing seasons of 2020 and 2021.

Systemic lupus erythematosus (SLE) and primary Sjögren's syndrome (pSS) share clinical as well as pathogenic similarities. Although previous studies suggest various abnormalities in different immune cell compartments, dedicated cell-type specific transcriptomic signatures are often masked by patient heterogeneity. Here, we performed transcriptional profiling of isolated CD4, CD8, CD16 and CD19 lymphocytes from pSS and SLE patients upon T cell stimulation, in addition to a steady-state condition directly after blood drawing, in total comprising 581 sequencing samples.

Objective: To determine whether adding obinutuzumab to standard-of-care lupus nephritis (LN) therapy could improve the likelihood of long-term preservation of kidney function and do so with less glucocorticoids.

Methods: Post hoc analyses of the phase II NOBILITY trial were performed. Time to unfavorable kidney outcome (a composite of treatment failure, doubling of serum creatinine, or death), LN flare, first 30% and 40% declines in estimated glomerular filtration rate (eGFR) from baseline, and chronic eGFR slope during the trial were compared between patients with active LN who were randomized to take obinutuzumab (n = 63) or placebo (n = 62) in combination with mycophenolate mofetil and glucocorticoids.

Background: Coronary microvascular dysfunction (CMD) and hypertension (HTN) occur frequently in hypertrophic cardiomyopathy (HCM), but whether blood pressure (BP) influences CMD and outcomes is unknown.

Objective: The purpose of this study was to test the hypothesis that HTN is associated with worse CMD and outcomes.

Methods: This retrospective study included 690 HCM patients.

Purpose: Myocardial perfusion (MP) stress single-photon emission computed tomography (SPECT) is an established diagnostic test for patients suspected of coronary artery disease (CAD). Meanwhile, coronary artery calcification (CAC) scoring obtained from diagnostic CT is a highly sensitive test, offering incremental diagnostic information in identifying patients with significant CAD yet normal MP stress SPECT (MPSS) scans. However, after decades of wide utilization of MPSS, CAC is not commonly reimbursed (e.

Oxaliplatin is a platinum-based alkylating chemotherapeutic agent used for cancer treatment. At high cumulative dosage, the negative effect of oxaliplatin on the heart becomes evident and is linked to a growing number of clinical reports. The aim of this study was to determine how chronic oxaliplatin treatment causes the changes in energy-related metabolic activity in the heart that leads to cardiotoxicity and heart damage in mice.
