The giant anterior salivary gland cells from the large mammalian blood-sucking, glossiphoniid leech, Haementeria ghilianii, can be subdivided into three morphologically and functionally distinct regions: 1) a soma, responsible for the synthesis and storage of secretory products; 2) a long cell process, responsible for the storage and intracellular transport of the secretory vesicles; and 3) the site of exocytosis at the process terminal. The giant somata are densely packed with secretory vesicles. Deep plasmalemmal invaginations invade the soma and form an extensive system of extracellular lacunae.

Chemotherapeutic evaluation using clinical criteria in spontaneous, autochthonous murine breast tumors.

Recent studies have shown that the dermis of scutate scales is involved in the expression of the beta keratin gene products, which characterize terminal differentiation of the epidermis on the outer scale surface (Sawyer et al.

Septic arthritis of the hip: a rare complication of angiography.

Local infection at the puncture site following percutaneous angiography has been reported as exceedingly rare. We describe the first patient with documented septic arthritis of the hip following percutaneous transfemoral arteriography. Radiologists should be aware of this potentially serious complication.

Unusual head and neck manifestations of non-Hodgkin's lymphomas in children and adults.

Certain patterns of "lymphoma masquerade" are examined, where the diagnosis is very difficult to establish. The presence of prolonged symptoms prior to the acute event or indolent slow healing wounds should alert clinicians to the possibility that lymphoma may be involved. Localizing tumors in order to make a tissue diagnosis is one potential problem encountered even with computerized tomography, but magnetic resonance imaging has been helpful in demonstrating the presence of tumor mass.

Intraperitoneal bladder rupture in a battered child.

Visceral manifestations of child abuse are less commonly diagnosed than are the characteristic skeletal injuries. We describe a child with intraperitoneal bladder rupture caused by blunt abdominal trauma. Intraperitoneal resorption of the extravasated urine led to laboratory evidence of renal failure.

Epithelial-mesenchymal interactions play important roles in the development of the vertebrate integument with its diverse appendages. As a result of these interactions, specific morphogenetic events occur which result in the formation of distinct epidermal appendages. Following the early morphogenetic events involving cell proliferation and movement, other developmental events such as stratification, histotypic differentiation, and terminal cytodifferentiation occur in the epidermis.

Reduction with age in the relaxation to beta-adrenoceptor agonists and other vasodilators in rat aorta.

We have examined the relaxation produced by the beta-adrenoceptor agonists isoprenaline and procaterol, and by sodium nitroprusside, forskolin and isobutylmethylxanthine in KCl (40 mmol/l)-contracted aortae from 6 week, 3 month and 6 month old Sprague-Dawley rats. The maximum relaxation to all agents was greatest in aortae from 6 week old animals. The relaxation to the beta-adrenoceptor agonists was virtually abolished, and the relaxation to the other agents significantly reduced, in aortae from 3 and 6 month old animals.

Morphogenesis and malformations of the skin NICHD/NIADDK research workshop.

Developmentally caused skin malformations constitute a spectrum of birth defects, some of which can be recognized prenatally by morphologic or biochemical means. The number of prenatally diagnosable skin diseases could be greatly expanded with an increased understanding of the molecular and cellular bases of skin development and the mechanisms that result in the generation of skin defects. The National Institute of Child Health and Human Development and the National Institute of Arthritis, Diabetes, Digestive and Kidney Diseases, therefore, sponsored a workshop that recommended basic biologic studies combined with clinical investigations of normal and abnormal cutaneous development set forth in this article.

Failure of L-histidinol to improve the therapeutic efficiency of 5-fluorouracil against murine breast tumors.

It has been reported that L-histidinol, a structural analogue of the essential amino acid L-histidine, can transiently inhibit proliferative cycling in cells with normal phenotype while allowing continued cell cycle transit in tumor cells. Thus, in the presence of L-histidinol, the toxicity of a proliferation-dependent drug such as 5-fluorouracil (FUra) was found to be reduced in normal tissue cells of the DBA/2J mouse, but not in L1210 leukemia cells in the same mouse. Because of the potential clinical significance of this approach to reduce chemotherapy-associated host toxicity, we evaluated the L-histidinol-FUra combination in a nonleukemic, solid murine tumor model, the BALB/c X DBA/8 F1 (hereafter called CD8F1) breast tumor.

Surface transformation of ram spermatozoa in uterine, oviduct and cauda epididymal fluids in vitro.

Genital tract fluids were collected continuously from conscious ewes through catheters inserted surgically into the uterus and oviducts. Cauda epididymal spermatozoa and fluid were obtained through catheters inserted into the transected vas deferens. The washed spermatozoa were labelled using the surface-specific chloroglycoluril-Na125I procedure.

The ontogeny of pulmonary alveolar macrophages in parabiotic mice.

A bilateral distribution of labeled blood monocytes was achieved by unilaterally labeling donor members of surgically parabiosed B6D2F1/J mice with tritiated thymidine (3HTdR). Labeled pulmonary alveolar macrophages (PAM) were found in lung lavage fluid of donors. By contrast, no labeled PAM were found in lung lavage fluid from unstimulated recipients, even 12 days after labeling.

Direct cardiac actions of phenylephrine when used in assessment of baroreflex function in the rat.

In pentobarbitone-anaesthetised wistar rats, the baroreflex bradycardia in response to the vasopressor agent phenylephrine was converted to a tachycardia by propranolol. In pithed wistar rats, phenylephrine produced a marked tachycardia which was mediated largely by beta-adrenoreceptors since propranolol markedly antagonised the response. A component of the tachycardia was mediated by alpha 1-adrenoreceptors since the tachycardia to phenylephrine in the presence of propranolol was antagonised by prazosin and mimicked by the alpha 1-adrenoreceptor agonist amidephrine.

Expression of beta keratin genes during skin development in normal and sc/sc chick embryos.

The expression of RNA sequences specific for scale beta (beta)-keratins has been followed during skin development in normal and scaleless (sc/sc) embryos. Total RNA from skin at various stages (36-46) of development, as well as newly hatched chicks, was immobilized on nitrocellulose paper and hybridized with a [32P]cDNA probe to beta-keratins (pCSK-12). Sequences for beta-keratins showed patterns of expression which were specific for each genotype and scale type examined.

Sialadenoma papilliferum of the adenoids in a 2-year-old child.

We report a case of sialadenoma papilliferum arising in the adenoids in a 2-year-old child. The tumor demonstrated papillary and adenomatous proliferations, suggestive of minor salivary gland origin. A review of the literature indicated no reported cases of this neoplasm arising in individuals this young.

Percutaneous transluminal angioplasty of the renal arteries: double-catheter technique.

A technique is described for angioplasty of the renal arteries using two catheters in a patient with two renal arteries to the left kidney. Injury to the nonstenotic supplemental artery during balloon dilatation of the stenotic main artery was avoided with the use of this technique.

A comparison of hydrocortisone and triamcinolone inhibition of scale and feather development.

Hydrocortisone (30-40 micrograms on day 10) and triamcinolone (10-20 ng on day 7-8) both inhibit or alter morphogenesis of scales and feathers. However, there are marked temporal and region-specific differences in the effects induced by these two glucocorticoids. Triamcinolone (TAC) is most teratogenic on day 7 or 8, inhibiting formation of spurs and feathers and inducing club feather formation.

Differences in the histogenesis and keratin expression of avian extraembryonic ectoderm and endoderm recombined with dermis.

The responses of the chorionic ectoderm and allantoic endoderm (from 8-day chick embryos) to dermal induction were compared through tissue recombinants grafted onto the chorioallantoic membrane. The chorionic epithelium formed the appropriate epidermis with a fully developed stratum corneum in response to both spur and scutate scale dermises. Analysis of these recombinant epidermal tissues by sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE) demonstrated that tissue-specific expression of the alpha (alpha) and beta (beta) keratin polypeptides occurred.

The cytokinetic behavior of pulmonary alveolar macrophages in monocytopenic mice.

The cytokinetic behavior of pulmonary alveolar macrophages (PAM) was studied by pulse labeling with 3HTdB in mice made monocytopenic by a single intravenous injection of the bone-seeking isotope strontium-89 (89Sr). In the presence or absence of blood monocytes, PAM population size was unchanged for up to 1 month of chronic, severe monocytopenia. Pulse-labeling studies performed during monocytopenia show that in control mice PAM population half-times were 17.

The significance of local resident pulmonary alveolar macrophage proliferation to population renewal.

The local proliferative capacity of resident pulmonary alveolar macrophages (PAM) was determined in normal CD-1 (ICR) mice and mice made severely monocytopenic with strontium-89 (89Sr). Intravenous injection of 2 muCi/gm BW 89Sr rendered mice monocytopenic for a 1-month period. During this time, in addition to there being no change in the size of the PAM population, flash 3HTdR labeling studies showed that at any one instant about 8.

Intermediate filament expression and lifespan potential in human somatic cell hybrids.

Limited lifespan human diploid fibroblast cells have been fused with the HeLa derived cell line HEB 7A which possesses transformed growth characteristics and unlimited division potential. HEB 7A expresses keratin intermediate filaments, while the fibroblast cells express only vimentin intermediate filaments. Independently arising clones of hybrids were examined for the presence of keratin by indirect immunofluorescence.
