Our laboratory recently made the novel observation that 5-hydroxytryptamine 1F (5-HT) receptor activation induces mitochondrial biogenesis (MB), the production of new, functional mitochondria, in vitro and in vivo. We sought to determine the mechanism linking the 5-HT receptor to MB in renal proximal tubule cells. Using LY344864 , a selective 5-HT receptor agonist, we determined that the 5-HT receptor is coupled to Gα and induces MB through Gβγ-dependent activation of Akt, endothelial nitric oxide synthase (eNOS), cyclic guanosine-monophosphate (cGMP), protein kinase G (PKG), and peroxisome proliferator-activated receptor-γ coactivator-1α (PGC-1α).
