Background: In children and adults with sickle-cell disease (SCD), acute ischemic stroke (AIS) associated with a vaso-occlusive crisis is a leading cause of physical and cognitive disability and death. However, neurological guidelines for acute management of AIS fail to directly address this issue. We here report a case of a man with severe cerebrovascular complications and illustrate the current evidence on the management of SCD-related AIS.

Article Synopsis
  • Deep brain stimulation of the subthalamic nucleus (STN-DBS) offers motor benefits for Parkinson's disease patients, but its effectiveness on postural and gait issues remains uncertain.
  • Physical therapy (PT) has shown effectiveness in improving mobility and stability in non-surgically treated Parkinson's patients, but its role after STN-DBS is less explored.
  • A study involving 21 experts highlighted that while PT could enhance motor function and quality of life post-STN-DBS, there’s a preference for conventional therapies over massage or manual techniques.

Background: The success of HER2-positive (HER2+) breast cancer treatment with trastuzumab, an antibody that targets HER2, relies on immune response. We demonstrated that TNFα induces mucin 4 (MUC4) expression, which shields the trastuzumab epitope on the HER2 molecule decreasing its therapeutic effect. Here, we used mouse models and samples from HER2+ breast cancer patients to unravel MUC4 participation in hindering trastuzumab effect by fostering immune evasion.
