Cellular target engagement technologies enable quantification of intracellular drug binding; however, simultaneous assessment of drug-associated phenotypes has proven challenging. Here, we present cellular target engagement by accumulation of mutant as a platform that can concomitantly evaluate drug-target interactions and phenotypic responses using conditionally stabilized drug biosensors. We observe that drug-responsive proteotypes are prevalent among reported mutants of known drug targets.

PLNC8 αβ is a cationic antimicrobial peptide that previously has been reported to express both antibacterial and antiviral properties. This study aimed to further elucidate the antiviral effects of PLNC8 αβ and its impact on virus-induced cytotoxicity and inflammatory signaling in human alveolar epithelial cells (A549) infected with the flavivirus Kunjin. Complementary in silico analyses using molecular dynamics (MD) simulation were conducted to investigate the mechanism of action of PLNC8 αβ by studying the interaction of PLNC8 α and β with models of a flavivirus membrane and a eukaryotic plasma membrane, respectively.

Background: Falls are a prevalent and serious concern among individuals in their older life. Residual impairments, such as balance and gait deficiencies, reduced sensory processing, and diminished strength, are the common causes of falls. Balance and mobility deficits can last for years and are linked to a high rate of falls (>6 months).

Article Synopsis
  • Disorders of consciousness (DOC) involve varying levels of awareness and arousal, and coma arousal therapies aim to improve these conditions, although their effectiveness is still being debated.
  • A meta-analysis of 260 studies narrowed down to 31 trials demonstrated that different forms of sensory stimulation significantly improve outcomes for DOC patients, particularly for those receiving treatment after 3 months of illness.
  • The findings indicate that sensory stimulation therapy is the most effective method among the interventions assessed, leading to improved behavioral responses in patients with DOC.

Biomedical research collaborations are to be contextualized in the larger global health agenda which also opens up new information pathways, expands research networks, and brings additional resources. A qualitative inquiry was employed to understand the perceived benefits and challenges of research collaborations by biomedical scientists from India (Global South [GS] country) and the Global North (GN). In-depth interviews were conducted with 47 biomedical scientists from India and 06 from the GN.

The mitotic MTH1 inhibitor TH1579 is a dual inhibitor that inhibits mitosis and incorporation of oxidative DNA damage and leads to cancer-specific cell death. The response to immune checkpoint inhibitor (ICI) treatment is often augmented by DNA damaging agents through the cGAS-STING pathway. This study investigates whether TH1579 can improve the efficacy of immune checkpoint blockades through its immunomodulatory properties.

Background Mucormycosis, attributed to a group of molds known as mucormycosis, is a rare yet life-threatening fungal infection often colloquially referred to as black fungus. While its incidence notably surged during the second wave of COVID-19 infections in India, it's essential to recognize that mucormycosis was a significant concern even before the advent of the pandemic. Understanding the prevalence and characteristics of this infection in the pre-COVID era provides a crucial context for evaluating its impact and dynamics during the pandemic.

Background The literature points towards the fact that paediatric elbow fractures happen more frequently and have greater variability when contrasted with adults. Between 65%-75% of pediatric fractures involve the upper extremity, and supracondylar humerus is the most common of them all. To know the exact site of injury and to estimate the degree of reduction after manipulation, the modified Baumann's angle, the Baumann's angle, the Humero-condylar angle, the Anterior humeral line, and the Radio-capitellar line are the parameters most commonly used.

In this study, an attempt was made to evaluate the relative efficacy of two important anti-gout agents, allopurinol and febuxostat, in the control of hyperuricaemia/gout using a poultry model. A 21-day study was conducted on 48 Vencobb-400 broiler chicks randomly divided into four groups. In one group hyperuricaemia/gout was induced by the oral administration of diclofenac (group D); in two other groups the ameliorative effect of the two drugs under study was investigated by providing both simultaneously, i.

Isolated Hoffa fractures of the femur are often missed on initial radiographic evaluations. Routine CT scans for intraarticular fractures in suburban populations are not routinely done. Nonunion of medial condyle Hoffa fracture of the distal femur is a rare finding and presents late with pain in weight-bearing and painful flexion.

Article Synopsis
  • Endothelin (ET)-1, a peptide from endothelial cells, causes a short-term increase in nitric oxide (NO) production followed by a decrease with sustained exposure, highlighting the complexity of its effects on endothelial function.
  • The study revealed that acute exposure to ET-1 boosts NO levels in pulmonary arterial endothelial cells (PAEC) by activating pp60 and promoting the phosphorylation of endothelial nitric oxide synthase (eNOS) at specific sites, which is influenced by caveolin-1 interactions.
  • Additionally, the research identified that inhibiting calcineurin can diminish the ET-1-induced increase in NO production by affecting the pp60-eNOS signaling pathway, suggesting potential therapeutic targets for enhancing vascular function.

Background:  Bone tumors remain a formidable challenge for orthopedic surgeons. In developing countries, the challenge is exacerbated by limited diagnostic, therapeutic, and management facilities and ignorance. Patients with upper and lower-extremity muscle and skeletal tumors are candidates for amputation or surgical rescue of the limbs.

Introduction Bone mineral density (BMD) is an essential indicator for diagnosing osteoporosis and evaluating the success of osteoporotic treatment. Dual-energy X-ray absorptiometry (DEXA), quantitative ultrasonography (QUS), and quantitative computed tomography (QCT) are frequently used for measuring BMD. The objective of the study was to evaluate the ability of QUS to screen for osteoporosis and bone density in postmenopausal women by calibrating it against DEXA.

Study of 35 days was conducted to evaluate citric acid (CA) as an additive in poultry broiler feed with lower mineral content of calcium (Ca) and total phosphorus (TP) in commercial broiler poultry birds for its effect on growth, nutrient utilization, carcass characteristics, and economics. Vancobb-400 strain day old broiler chicks were divided into four main treatment groups T T, T, and T Treatment groups were further divided into eight replicates with ten chicks in each. T served as control, given standard corn-soy flakes-based ration (Pre-starter %: Crude protein (CP)-23, Ca-1.

This paper re-examines the causality between stock returns and foreign portfolio investment (FPI) flows in the Indian context during the COVID-19 pandemic. Using the Covid-19 index constructed by Narayan et al. [19] and the Toda and Yamamoto Granger causality test, the study reveals that bi-directional causality runs from FPI flows to stock returns in the early period of the Covid-19 pandemic.

This study reassesses the dynamic relationship between stock, oil, foreign exchange markets, and COVID-19-induced uncertainty. For estimation, we utilize the monthly data from January 2020 to December 2021 and utilize panel vector autoregression econometric techniques in emerging market economies. The empirical findings reveal that heightened uncertainty during the pandemic had a negative impact on stock and oil, and foreign exchange markets.
