The native extracellular matrix is continuously remodeled to form complex interconnected network structures that reversibly regulate stem cell behaviors. Both regulation and understanding of its intricate dynamicity can help to modulate numerous cell behaviors. However, neither of these has yet been achieved due to the lack of designing and modeling such complex structures with dynamic controllability.

Introduction: It remains unclear how patients with atopic dermatitis (AD) and clinicians perceive the level of patient-clinician communication and if there could be potential lapses. This cross-sectional study aims to compare perspectives between patients with AD and dermatologists regarding communication and treatment expectations in Asia.

Methods: Moderate-to-severe patients with AD and practicing dermatologists were recruited from eight Asia-Pacific territories, including Mainland China, Hong Kong, India, Japan, Singapore, South Korea, Taiwan, and Thailand.

Article Synopsis
  • Mesenchymal stem cells (MSCs) have potential in medical therapies due to their ability to modulate immune responses, and this study focused on their use in treating atopic dermatitis (AD).
  • A phase 1 trial confirmed the safety of allogenic human bone marrow-derived clonal MSCs (hcMSCs) in patients, leading to a phase 2 trial that showed hcMSC treatment resulted in improved eczema severity after 12 weeks compared to a placebo.
  • Despite some mild adverse effects, hcMSC therapy had an acceptable safety profile, suggesting it could be effective, though further extensive research is needed to validate these initial findings.

  • Cyclosporine (CS) is commonly used to treat moderate to severe atopic dermatitis (AD), but its long-term effects on blood pressure are not well understood.!
  • A study analyzed data from over 1.8 million AD patients to examine how CS dosage affects the risk of hypertension, finding that current CS use significantly increases this risk.!
  • While low-dose CS has a low incidence of hypertension, higher dosages and longer usage are linked to a greater likelihood of developing high blood pressure, suggesting caution is needed for long-term prescriptions.!

Background: Atopic dermatitis (AD) is a common skin disease which, depending on its severity, can have a significant impact on the quality of life of affected individuals. In cases of severe AD, systemic immunomodulatory agents can be considered for treatment. However, the available treatment options for moderate AD are limited.

  • Psoriasis is a challenging condition that significantly affects patients' well-being, and the psychological effects of its treatments are often overlooked.
  • This study aimed to evaluate the impact of the medication adalimumab on health-related quality of life in Korean patients with psoriasis over a 24-week period, using various patient-reported outcome measures.
  • Results showed significant improvements in quality of life scores and treatment satisfaction, indicating that adalimumab is effective and well-tolerated for managing psoriasis in these patients.

Background: Data illustrating the impact of atopic dermatitis (AD) on lives of adults with AD in South Korea are limited.

Objective: To assess the AD disease severity and its impact on quality of life (QoL) in patients with AD from South Korea.

Methods: Patients with AD utilizing the specialist dermatology services of major hospitals in South Korea were assessed for disease severity using Eczema Area and Severity Index (EASI) score, for QoL using Dermatology Life Quality Index (DLQI) (for QoL), and for comorbidities and treatment experience via retrospective review of 12-month medical records.

Skin aging is a major risk factor for the dermal diseases, and interventions to attenuate cellular senescence are expected to reduce the risk for age-related diseases involving skin atrophy. However, blocking cell death or extending proliferation causally results in side effects and an increased cancer risk. For identification of a safer approach, we focused on PDK1 inhibition, which could revert cellular senescence and reduce senescence factors in skin in vitro, in a human skin equivalent model and in an exploratory, placebo-controlled, interventional trial.

The phenotypes of atopic dermatitis (AD) are diverse, and ethnic differences have been suggested. To date, few studies have explored large-scale national data on the treatment patterns of AD in Asians. Therefore, we aimed to examine real-world treatment patterns for AD, including the probability of discontinuation of AD treatment and restart after discontinuation.

Background: In psoriasis treatment, not all body regions improve simultaneously after clinical interventions.

Objective: This study was aimed at evaluating clinical responses across body regions, which may differentially influence patient treatment plans.

Methods: This prospective, observational, and multi-center study was conducted in Koreans who adhered to ustekinumab treatment based on criteria per local label and reimbursement guidelines.

Environmentally hazardous substances and exposure to these can cause various diseases. Volatile organic compounds can easily evaporate into the atmosphere, thereby exerting toxic effects through either the skin or respiratory tract exposures. Toluene, a neurotoxin, has been widely used in various industries.

Skin barrier dysfunction induces skin inflammation. Signal transducer and activator of transcription 3 (STAT3) is known to be involved in Th17-mediated immune responses and barrier integrity in the cornea and intestine; however, its role in the skin barrier remains largely unknown. In this study, we elucidated the potential role of STAT3 in the skin barrier and its effect on kallikrein-related peptidase 5 (KLK5) and serine protease inhibitor Kazal-type 5 (SPINK5) expression using a mouse model with keratinocyte-specific ablation of STAT3.

Background: Hand eczema refers to eczema located on the hands, regardless of its etiology or morphology. Despite its high prevalence and significant impact on patients' quality of life, treatment is frequently challenging because of its heterogeneity, chronic and recurrent course, and lack of well-organized randomized controlled trials of the various treatment options.

Objective: These consensus guidelines aim to provide evidence-based recommendations on the diagnosis and management of hand eczema to improve patient care by helping physicians make more efficient and transparent decisions.

Hypoxia-inducible factor-1α (HIF-1α) has been reported to be up-regulated in psoriatic epidermis, resulting in increased proliferation and abnormal differentiation of human keratinocytes (KCs). However, the role of HIF-1α in psoriatic epidermis, which is mainly composed of KCs, is poorly understood. Here, we show that morphogenic protein 6 (BMP6) is down-regulated when HIF-1α is upregulated in patients with psoriasis skin lesions.

Zika virus (ZIKV) has emerged as a global pathogen causing significant public health concern. ZIKV infections in humans principally occur via mosquito bites. Thus, host skin cells are permissive to ZIKV infection and are the first line of defense against the virus.
