Article Synopsis
  • * The study proposes that changes in the intracellular domain (ICD) of these precursors, triggered by certain signals, enhance the sensitivity of substances like neuregulin-1 and CD44 to proteolytic cleavage by enzymes such as chymotrypsin or ADAMs.
  • * The findings indicate that this regulation involves specific interactions between substrates and suggests a potential universal mechanism that could be applied to many different ADAM substrates, controlling their release in biological processes.

Ectodomain cleavage by A-disintegrin and -metalloproteases (ADAMs) releases many important biologically active substrates and is therefore tightly controlled. Part of the regulation occurs on the level of the enzymes and affects their cell surface abundance and catalytic activity. ADAM-dependent proteolysis occurs outside the plasma membrane but is mostly controlled by intracellular signals.
