Unlabelled: Glioblastoma (GBM) (grade IV glioma) is the most fatal brain tumor, with a median survival of just 14 months despite current treatments. Temozolomide (TMZ), an alkylating agent used with radiation, faces challenges such as systemic toxicity, poor absorption, and drug resistance. To enhance TMZ effectiveness, we developed poly(ethylene glycol) (PEG) liposomes co-loaded with TMZ and O6-benzylguanine (O6-BG) for targeted glioma therapy.

Cryopreservation of testicular tissue holds an important role in the field of fertility preservation, particularly for prepubertal boys diagnosed with cancer. However, prepubertal testicular tissue cryopreservation is still considered to be in the experimental stage necessitating the refinement of cryopreservation protocol. Considering the fact that loss of membrane lipids is the primary cause of freeze-thaw-induced loss of testicular cell functions, in this study, we explored the beneficial properties of exogenous supplementation of membrane lipids in the form of liposomes in enhancing the cryosurvival of prepubertal testicular tissue.

Aims: Development and characterization of LAM and DTG loaded liposomes conjugated anti-CD4 antibody and peptide dendrimer (PD2) to improve the therapeutic efficacy and to achieve targeted treatment for HIV infection.

Main Methods: A 2-level full factorial design was used to optimize the preparation of dual drug loaded liposomes. Optimized dual drug loaded ligand conjugated liposomes were assessed for their cytotoxicity and cell internalization on TZM-bl cells.

Clethodim is a widely used and approved class II herbicide, with little information about its impact on the reproductive system. Herein, we investigated the male reproductive toxicity of clethodim using a mouse model. GrassOut Max (26% clethodim-equivalent) or analytical grade clethodim (≥90%) were given orally to male mice for 10 d in varying doses.

To develop a new sensitive RP-HPLC method for simultaneous estimation of 5-fluorouracil (5-FU) and sonidegib (SDG). Analytical and bioanalytical methods for simultaneous quantification of 5-FU and SDG in bulk, nanoformulations and in rat plasma were developed and validated using a gradient elution technique. Separation of the analytes was effected on a Luna C18 LC column using a mobile mixture comprising acetonitrile and acidified water.

A novel isocratic stability-indicating chromatographic method was developed, optimized and validated using Design-Expert® following ICH guidelines for the quantification of Timolol maleate (TM). The intrinsic stability of TM was assessed by force degradation studies, which concluded no extensive degradation except under alkaline and oxidative conditions. TM was quantified accurately in the surfactant-based elastic vesicular system by separating it on Hypersil BDS C8 column using triethylamine in H2O (0.

Article Synopsis
  • A new reversed-phase high-performance liquid chromatography (HPLC) method has been developed to simultaneously measure dolutegravir sodium and lamivudine in a special drug formulation called nanoliposomes.
  • The method was optimized using a statistical approach called Box-Behnken design, focusing on factors like flow rate and organic phase ratio to improve accuracy in results like retention time and peak area.
  • Validation of the method followed international guidelines, confirming its ability to specifically detect both drugs even among other substances, allowing the researchers to assess how much of each drug is effectively loaded in the liposomal formulation.

Article Synopsis
  • A reverse phase high-performance liquid chromatography method was created and validated for measuring apremilast (APL) and betamethasone dipropionate (BD) in both bulk materials and drug-loaded microsponges.
  • The method involved optimizing conditions such as column temperature and flow rate through a central composite design, and it successfully demonstrated stability of both drugs under various stress conditions.
  • Results showed high levels of accuracy and precision with very low relative standard deviations, making the method suitable for quantifying APL and BD effectively in different formulations.

Article Synopsis
  • Prepubertal Swiss albino mice of both sexes were given first-line anti-tuberculosis drugs (ATDs) for 4 weeks to assess the impact on reproductive health.
  • The study found that ATDs significantly reduced sperm quality in male mice and negatively affected oocyte quantity and health in female mice, leading to various abnormalities.
  • Overall, the findings indicate that ATDs have detrimental effects on male and female gametes, though the degree of toxicity varies among the drugs.
