Use of glucagon-like polypeptide 2 analogs for intestinal failure.

Introduction: Over a half century ago, a component of glucagon was found to have potent gastrointestinal effects. Shortly after proglucagon was sequenced, its component peptides were characterized, and glucagon-like polypeptide-2 (GLP-2) was noted to have the most potent intestinotrophic properties improving fluid and electrolyte balance in experimental animals and humans.

Areas Covered: Glucagon-like polypeptide-1 (GLP-1) slows small intestinal motility more effectively, but its intestinotrophic properties are weaker.

Morphology and line tension of twist disclinations in a nematic liquid crystal.

We deploy optical microscopy with and without an applied magnetic field to characterize the three-dimensional morphology and measure the line tension of twist disclinations in twisted nematic liquid crystal (LC) sample cells. Twist disclinations are generated by quenching the LC, 5CB (4-cyano-4'-pentylbiphenyl), into the nematic phase; 5CB is confined between substrates with in-plane anchoring directions perpendicular to one another. The disclinations form loops separating domains of opposite twist handedness.

Tuning Chemical DNA Ligation within DNA Crystals and Protein-DNA Cocrystals.

Biomolecular crystals can serve as materials for a plethora of applications including precise guest entrapment. However, as grown, biomolecular crystals are fragile in solutions other than their growth conditions. For crystals to achieve their full potential as hosts for other molecules, crystals can be made stronger with bioconjugation.

An automatic glucose monitoring system based on periplasmic binding proteins for online bioprocess monitoring.

Glucose is one of the most vital nutrients in all living organisms, so its monitoring is critical in healthcare and bioprocessing. Enzymatic sensors are more popular as a technology solution to meet the requirement. However, periplasmic binding proteins have been investigated extensively for their high sensitivity, enabling microdialysis sampling to replace existing complex and expensive glucose monitoring solutions based on enzymatic sensors.

Red cell abnormalities characterized by ektacytometry in children with cholestasis.

Background: Spur-cell anemia sometimes accompanies cholestasis. We postulated that even in the absence of spur-cells, cholestasis might alter red blood cell (RBC) osmotic fragility and deformability. Therefore, we assessed these RBC measures by ektacytometry in pediatric patients.

Magnetic-field-driven director configuration transitions in radial nematic liquid crystal droplets.

We study the director configurations of nematic liquid crystal (NLC) droplets with homeotropic anchoring in a magnetic field and report observation of a magnetic-field-driven transition from a deformed radial to an axial-with-defect configuration. Magnetic-field-induced transitions in NLC droplets differ fundamentally from the traditional planar Freedericksz transition due to the spherical droplet geometry and resulting topological defect. This transition has been studied theoretically, but the director configurations and mechanism of defect evolution in an applied magnetic field have yet to be observed experimentally.

Modular Protein-DNA Cocrystals as Precise, Programmable Assembly Scaffolds.

High-precision nanomaterials to entrap DNA-binding molecules are sought after for applications such as controlled drug delivery and scaffold-assisted structural biology. Here, we engineered protein-DNA cocrystals to serve as scaffolds for DNA-binding molecules. The designed cocrystals, isoreticular cocrystals, contain DNA-binding protein and cognate DNA blocks where the DNA-DNA junctions stack end-to-end.

Plasma, urine, and stool metabolites in response to dietary rice bran and navy bean supplementation in adults at high-risk for colorectal cancer.

Introduction: Dietary intake of whole grains and legumes and adequate physical activity (PA) have been associated with reduced colorectal cancer (CRC) risk. A single-blinded, two-arm, randomized, placebo-controlled pilot trial was implemented to evaluate the impact of a 12-week dietary intervention of rice bran + navy bean supplementation and PA education on metabolite profiles and the gut microbiome among individuals at high risk of CRC.

Methods: Adults (n=20) were randomized 1:1 to dietary intervention or control.

Pilot study of multi-gene pharmacogenetic testing for pain management in oncology palliative medicine.

We evaluated the application and clinical impact of multi-gene pharmacogenetic testing in oncology palliative medicine. In a single-arm pilot trial, cancer patients with uncontrolled pain were assessed in a palliative medicine clinic at baseline and received pharmacogenetic testing. Results were used as applicable up to the final visit (day 30).

Correlation of Probe-Based Confocal Laser Endomicroscopy (pCLE) and Mucosal Integrity Testing (MIT) with Epithelial Barrier Function and Presence of Gastroesophageal Reflux Disease (GERD).

Introduction: Epithelial barrier function (EBF) disruption is a key mechanism underlying gastroesophageal reflux disease (GERD). Our aim was to assess whether two novel technologies, probe-based confocal laser endomicroscopy (pCLE) and mucosal integrity testing (MIT), could assess EBF.

Methods: We prospectively enrolled patients undergoing upper endoscopy for refractory GERD or non-GERD conditions.

Pain Management Using Clinical Pharmacy Assessments With and Without Pharmacogenomics in an Oncology Palliative Medicine Clinic.

Purpose: Approximately 30% of patients with cancer who have pain have symptomatic improvement within 1 month using conventional pain management strategies. Engaging clinical pharmacists in palliative medicine (PM) and use of pharmacogenomic testing may improve cancer pain management.

Methods: Adult patients with cancer with uncontrolled pain had baseline assessments performed by PM providers using the Edmonton Symptom Assessment Scale.

World Health Organization's Growth Reference Overestimates the Prevalence of Severe Malnutrition in Children with Sickle Cell Anemia in Africa.

Anthropometric indices are widely used to assess the health and nutritional status of children. We tested the hypothesis that the 2007 World Health Organization (WHO) reference for assessment of malnutrition in children with sickle cell anemia (SCA) overestimates the prevalence of severe malnutrition when compared to a previously constructed SCA-specific reference. We applied the WHO and SCA-specific references to children with SCA aged 5-12 years living in northern Nigeria (Primary Prevention of Stroke in Children with SCA in sub-Saharan Africa (SPRING) trial) to determine the difference in prevalence of severe malnutrition defined as body mass index (BMI) -score <-3 and whether severe malnutrition was associated with lower mean hemoglobin levels or abnormal transcranial Doppler measurements (>200 cm/s).

Learning hierarchically organized science categories: simultaneous instruction at the high and subtype levels.

Background: Most science categories are hierarchically organized, with various high-level divisions comprising numerous subtypes. If we suppose that one's goal is to teach students to classify at the high level, past research has provided mixed evidence about whether an effective strategy is to require simultaneous classification learning of the subtypes. This past research was limited, however, either because authentic science categories were not tested, or because the procedures did not allow participants to form strong associations between subtype-level and high-level category names.

Depressive Symptoms Predict Anti-tumor Necrosis Factor Therapy Noncompliance in Patients with Inflammatory Bowel Disease.

Background: Noncompliance in use of anti-tumor necrosis factor (anti-TNF) therapy in patients with moderate-to-severe inflammatory bowel disease (IBD) can be a factor in medication failure. Few studies have evaluated the contribution of depressive symptoms to medication noncompliance in anti-TNF therapies.

Methods: A retrospective chart review was performed in a single-center tertiary care IBD center for patients with Crohn's disease and ulcerative colitis starting anti-TNF therapy over a 2-year period.
