Lewis lung carcinoma cells from tumors, metastasis nodules, or from culture bind fluorescent derivatives of neoglycoproteins containing alpha-D-glucose residues: This binding is competitively inhibited by neoglycoproteins containing alpha-D-glucose, by mannan, and by several other neoglycoproteins. Cell binding and uptake of the fluorescent derivatives of the neoglycoproteins was quantified by lysing the cells with an alkylpolyol (MAC 19 or MAC 18) and measuring the fluorescence intensity of the supernatant. The mannose-specific adherence to yeast cells of Escherichia coli excreted in the urine of patients with urinary tract infections was compared with that of isolates from the same urine after growth of the bacteria in broth. Previous methods for the dissociation of pili caused their irreversible denaturation. We report the successful coreconstitution of solubilized lymphocyte plasma membranes and Sendai virus envelopes into membrane vesicles that possess the ability to fuse efficiently with mouse splenocytes of thymocytes to give fully viable cells with modified surface properties. Integration of donor membrane components into the acceptor cells was demonstrated by chemical, morphological, and immunological methods. Over 40% of the label from vesicles coreconstituted with radiolabeled (with 125I or 3H) lymphocyte membranes was incorporated into the fused cells.
