"Active" reservoir cells transcribing HIV can perpetuate chronic inflammation in virally suppressed people with HIV (PWH) and likely contribute to viral rebound after antiretroviral therapy (ART) interruption, so they represent an important target for new therapies. These cells, however, are difficult to study using single-cell RNA-seq (scRNA-seq) due to their low frequency and low levels of HIV transcripts, which are usually not polyadenylated. Here, we developed "HIV-seq" to enable more efficient capture of HIV transcripts - including non-polyadenylated ones - for scRNA-seq analysis of cells from PWH.

Objective: To identify the prevalence and factors associated with T-score discordance between the spine and hip, as well as between the paretic and non-paretic hips in hemiplegic stroke patients, this study investigated bone mineral density (BMD) patterns. Bone loss predominantly affects the paretic hip after a stroke, and typical clinical assessments using dual-energy X-ray absorptiometry (DXA) that scan the lumbar spine (LS) and a single hip may overlook an osteoporosis diagnosis. This oversight could potentially lead to suboptimal treatment for stroke patients.

Article Synopsis
  • Phytoplankton composition and biomass were studied in the C-43 Canal of southwest Florida, which receives regulated discharges from Lake Okeechobee, revealing dominance of harmful cyanobacteria during high discharge periods in spring and summer.
  • Conversely, during low discharge periods in mid-summer and autumn, phytoplankton biomass decreased, leading to increased dinoflagellate populations, with notable first records of Dinophysis baltica and Parvodinium goslaviense in Florida.
  • The findings suggest that managing water discharge rates can significantly impact phytoplankton dynamics and help mitigate harmful algal blooms in the canal and its downstream coastal areas.

Despite antiretroviral therapy (ART), HIV persists in latently-infected cells (the HIV reservoir) which decay slowly over time. Here, leveraging >500 longitudinal samples from 67 people living with HIV (PLWH) treated during acute infection, we developed a mathematical model to predict reservoir decay from peripheral CD4 + T cells. Nonlinear generalized additive models demonstrated rapid biphasic decay of intact DNA (week 0-5: t ~ 2.

With the increase in consumer interest in food safety, in this study, we aimed to investigate the antibacterial effect of peraceic acid (A, B, and Daesung; 50-200 ppm) and sodium hypochlorite disinfectants on chicken carcasses and contaminated water, respectively, and changes in the appearance of chicken carcasses. Considering the antibacterial effect of each disinfectant concentration, the most significant antibacterial efficacy was observed for general bacteria and at 200 ppm regardless of disinfectant type. Considering the disinfectant type at 200 ppm, sodium hypochlorite was the least effective, and peracetic acid A showed the highest antibacterial efficacy at all concentrations.

Article Synopsis
  • Fourier ptychography (FP) is a computational imaging technique that enhances image resolution and obtains phase information by using varied angles of light, but reconstructing images can be challenging, especially with noisy or incomplete data.
  • To address these challenges, a new algorithm combining anisotropic total variation and Tikhonov regularizations is proposed, improving image reconstruction quality in high-throughput scenarios.
  • The method, which utilizes the alternating direction method of multipliers, has been tested against other FP algorithms and shows promising results in noise reduction and preserving key details in images collected from biological specimens, even with sparse data.

Preexisting anti-interferon-α (anti-IFN-α) autoantibodies in blood are associated with susceptibility to life-threatening COVID-19. However, it is unclear whether anti-IFN-α autoantibodies in the airways, the initial site of infection, can also determine disease outcomes. In this study, we developed a multiparameter technology, FlowBEAT, to quantify and profile the isotypes of anti-severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) and anti-IFN-α antibodies in longitudinal samples collected over 20 months from the airways and blood of 129 donors spanning mild to severe COVID-19.

Pistachio shell powder is a high-fiber co-product from the pistachio nut industry that may provide energy and nutrients in animal diets, but no data have been reported for the nutritional value of pistachio shell powder when fed to pigs. Two experiments were, therefore, conducted to test the hypothesis that apparent total tract digestibility () of gross energy (), dry matter (), and total dietary fiber () and concentration of digestible energy () in pistachio shell powder are not different from those in soybean hulls when fed to gestating or lactating sows. In experiment 1, 24 gestating sows were housed in metabolism crates and fed a corn-based basal diet or 2 diets that contained corn and 20% pistachio shell powder or corn and 20% soybean hulls.
